Cutaneous scarring happens as significant amounts of fibroblasts differentiate into myofibroblasts during the repair process of skin injuries,which would accelerate the contraction of wounds and increase the deposition of collagens.In the current study,the synergistic effects of rBM-MSCs and PCL/Gelatin fibrous scaffolds on the differentiation of fibroblasts were investigated.The main works of this research include: In this research,the effects of combined rBM-MSCs and PCL/Gelatin fibrous scaffolds on the differentiation of fibroblasts were investigated.The main work of this research would be:(1)Co-culture of fibroblasts with rBM-MSCs via transwell system to explore the expression level of the differentiation associated gene of fibroblasts and rBM-MSCs with or without the stimulation of TGF-?1.(2)Co-culture of fibroblasts with rBM-MSCs on PCL/Gelatin fibrous scaffold via transwell system to explore the expression level of the differentiation-associated genes of fibroblasts and rBM-MSCs with or without the stimulation of TGF-?1.(3)Exploration of the effect with the conditional medium of PCL/Gelatin fibrous scaffold-treated rBM-MSCs on the differentiation of fibroblasts with or without the stimulation of TGF-?1.(4)Assessment of the effect of the combinational treatment(rBM-MSCs plus PCL/Gelatin fibrous scaffold)on scar formation by utilizing rat full-thickness excisional wound model.This study demonstrated that rBM-MSCs alone or rBM-MSCs seeded on PCL/Gelatin fibrous scaffold can alleviate the differentiation of fibroblasts into myofibroblasts,and the latter therapy was more advantageous.The advantage was further confirmed in full-thickness excisional wound model.rBM-MSCs affected the differentiation of fibroblasts by secreting cytokines,which was promoted by introducing PCL/Gelatin fibrous scaffolds.Taken together,these results proved that combinational treatment(rBM-MSCs plus PCL/Gelatin fibrous scaffold)attenuated the differentiation of fibroblasts and reduced scar formation.
