Objective:To observe the effect of wrist ankle acupuncture on pain behavior,the expression of glutamate and NMDA receptor phosphorylation protein in spinal dorsal horn of rats with neuralgia.Methods:A total of 36 SD rats were randomly divided into sham group,model group,WA group,with12 rats in each group.The SNI rats model was made by cutting the common peroneal nerve and tibial nerve in the sciatic nerve after ligation with 5-0 silk thread and preserving the sural nerve.The success criteria of the model were the decrease of the mechanical pain threshold and the increase of the duration of cold stimulation.In the sham operation group,only the sciatic nerve was exposed,the common peroneal nerve and tibial nerve were not ligated,and other operations such as model group were performed.The WA group received“R4”-“R5”-“R6”points for treatment from the 5th day to the 14th day after modeling.In the sham operation group and the model group,there was no intervention.On the 5th and 14th day after operation,the pain behavior of mechanical pain was recorded by von Frey pain detector,the pain behavior of cold stimulation was recorded by acetone,and the glutamate content was detected by 1H-MRS.On the 14th day after operation,the glutamic acid content in spinal dorsal horn was detected by ELISA,and the expression of NMDAR1and NMDAR2 phosphorylated protein was detected by immunohistochemistry.All data were processed by spss20.0 statistical software.Results:?1?Mechanical pain behavior:compared with the sham operation group,the mechanical pain threshold of the model group was significantly lower on the 5th,10th and14th day after operation?P<0.01?;compared with the model group,the mechanical pain threshold of the wrist ankle acupuncture group was significantly higher on the 14th day after operation?P<0.01?.?2?Behavioral study of cold stimulation pain:compared with the sham operation group,the duration of cold stimulation foot contraction in the model group increased significantly on the 5th,10th and 14th day after operation?P<0.01?;compared with the model group,the duration of cold stimulation foot contraction in the wrist ankle acupuncture group decreased significantly on the 14th day after operation?P<0.01?.?3?Magnetic resonance spectroscopy detection:compared with the sham operation group,the Glu/Cr ratio in the model group increased significantly on the 5th and 14th day?P<0.01?,and the Glu/Cr ratio in the wrist ankle acupuncture group increased significantly on the 5th day?P<0.01?;compared with the model group,the relative expression of Glu/Cr in the wrist ankle acupuncture group decreased on the 14th day?P<0.01?.?4?Enzyme linked immunosorbent assay:compared with the sham operation group,the concentration of glutamate in the model group was significantly higher?P<0.05?;compared with the model group,the concentration of glutamate in the wrist ankle acupuncture group showed a downward trend,the difference was statistically significant?P<0.05?.?5?Immunohistochemical test:on the 14th day after operation,compared with the sham operation group,the expression of NMDAR1 and nmdrar2 phosphorylated protein in the model group increased?P<0.01?;compared with the model group,the expression of NMDAR1 and NMDAR2 phosphorylated protein in the wrist ankle acupuncture group decreased significantly?P<0.01?.Conclusion:?1?Wrist ankle acupuncture can improve the abnormal behavior pattern of sciatica rats,and inhibit the expression of glutamate and NMDAR1,NMDAR2phosphorylation protein in spinal dorsal horn.?2?Wrist ankle acupuncture may play an analgesic role by reducing the contents of glutamate and NMDA receptor in spinal dorsal horn of sciatica rats and inhibiting the excitability of nociceptive neurons.
