Abstract/Summary:
Objective : Intervertebral disc degeneration(IVDD)is a common and frequently-occurring disease in middle-aged and elderly people.Neck pain and low back pain is the main symptom.At present,the treatment mainly involves conservative treatment such as physiotherapy and medicine,and TLIF surgery.Although its clinical efficacy is acceptable,some patients still have poor results.The main reason is that the pathogenesis of the disease is not well understood.Long non-coding RNAs are a class of non-coding RNAs more than 200 nt in length,which have important biological functions in physiological and pathophysiological processes.However,its role in IVDD and its molecular mechanism are in its infancy and less studied.This experiment hopes to study the differentially expressed LncRNAs in IVDD nucleus tissue by high-throughput sequencing,explore the molecular mechanism of LncRNA regulation of IVDD,and provide a target for the treatment of IVDD.Method:High-throughput screening and bioinformatics analysis was used to research differential expression of LncRNAs in IVDD and;Linc01776 knockout plasmid vector was constructed and study of Linc01776 biologic function in IVDD was performed;competition endogenous RNA(CeRNA)regulatory mechanisms of the Linc01776 was predicted.Result:Through analysis of RNA-seq data,a total of 23,706 differentially expressed LncRNAs were obtained,of which 233 were up-regulated by more than 2 fold change and 340 were down-regulated by more than 2 fold change.Linc01776 is the most prominent differential fold in up-regulated lncRNAs,which play important biological functions in IVDD.CeRNA regulatory mechanisms of Linc01776/miR-196b-5p/Akt1 was predicted by bioinformatics.Conclusion:LncRNAs is involved in the occurrence and development of IVDD.Linc01776 is the most significant difference in up-regulated lncRNAs,suggesting that this LncRNA may play an important biological role in IVDD.Linc01776/miR-196b-5p/Akt1 may be a mechanism for its regulation of IVDD progression.
