Background and objective:hn RNP K protein is closely related to many oncogenes and growth factors.A large number of studies have found that mi RNAs are involved in the occurrence and development of gastric cancer as anti-oncogenes or proto-oncogenes.This study explores the relationship between mi RNAs and hn RNP K,and aims to provide new insights into the mechanism of mi RNAs and hn RNP K in gastric cancerMethods:1.The expression of hn RNP K in normal gastric tissues and gastric cancer tissues were detected by immunohistochemistry and Western Blot.The expression profiles of mi RNAs in gastric cancer cells with high expression of hn RNP K were screened by microarray.The expression profilewas verified by Real time q-PCR real-time.2.Data of gastric cancerm RNA and expression of mi RNAs were collected through TCGA database to screen the differentially expressed mi RNAs corresponding to hn RNP K overexpression.The co-expressed mi RNAs were analyzed by interaction between TCGA database and chip analysisdata,and verified by Real time q-PCR.3.Construction of hn RNP K overexpression and knockdowncells,Western Blot was used to confirm the effect of overexpression and knockdown,and Real time q-PCR was used to verify the expression of screened mi R-20 a.Results:1.High expression of hn RNP K protein was found in gastric cancer tissues and cells.Fourteen kinds of mi RNAs were up-regulated and down-regulation of seven in gastric cancer cells with high expression of hn RNP K.Thedata of Real time q-PCR showed that 13 mi RNAs were up-regulated by more than two times.2.The co-expressed mi RNAs(mi R-17/20a/24/182)were obtained by the interaction analysis of TCGA database and the cell chip.The detection in tissues by Real time q-PCR showed that only mi R-20a/182 were up-regulated.3.The hn RNP K overexpression and knockdown cell were successfully constructed.The Real time q-PCR data showed that only mi R-20 a was significantly up-regulated in the gastric mucosal cells with hn RNP K overexpression;while the endogenous hn RNP-K protein was knocked down,mi R-20 a was significantly down-regulated;TCGA data analysis showed that hn RNP K gene was correlated with mi R-20 a in gastric cancer,and mi R-20 a was up-regulated in gastric cancer.Conclusions:1.hn RNP K was highly expressed in gastric cancer tissues and cells,and mi RNA was co-expressed by hn RNP K in gastric cancer cells.2.It was found that hn RNP K protein could up-regulate the expression of mi R-20 a in gastric cancer cells.
