DNA flowers fabricated by rolling circle amplification(RCA)can be used to act as carriers for various biomedical applications.The fully understanding of the intracellular location and stability of DNA flowers in live cells is urgently demanded.Here,we co-assembled DNA flowers with RIR fluorophores,which have the properties of aggregation-induced emission(AIE).We use the RIR-DNA flowers for direct visualization of distribution and degradation of DNA flowers in live cellular systems.The results show DNA flowers entered live cells and accumulated in lysosome,and dissociated within 72 hours.And RIR-DNA flowers can be used as fluorophores for specific detection of lysosomal in cell imaging.Photothermal therapy(PTT)is an emerging therapeutic strategy for cancer treatment,but single PTT has many limitations.We fabricated AuNRs-Dox-DNA flowers with tumour-specific targeting effect(MADF)by One-Pot Synthesis.AuNRs are photosensitizers for PTT,Dox is a typical kind of chemotherapy drug,MUC-1 aptamer has tumour-specific targeting effect.The results show MADF has the ability of photothermal conversion;The cellular internalization ability of MADF is better than free AuNRs,free Dox and AuNRs-Dox-DNA flowers without tumour-specific targeting.But the therapeutic efficacy of MADF is as same as free AuNRs and free Dox for MCF-7.Therefore,we will investigate it in multidrug resistant MCF-7 and further verify in tumor-bearing nude mice.
