Cancer,known as malignant tumor,is a complex disease caused by a variety of factors and is a common serious disease that directly endangers human life in today's world.The uncontrolled growth of cancer cells is the main cause of the disease,and cancer cells continue to spread,invading other organs of the body and causing exhaustion,eventually leading to death.Obviously,the damage that cancer poses to human health is a very serious problem.Breast cancer is the most common malignant tumor in women and one of the leading causes of cancer-related deaths in women.The life and health of women is seriously threatened worldwide.The latest Globocan 2018 data reports that both morbidity and mortality are among the top cancer related deaths among women,and breast cancer patients are becoming younger.Young breast cancer patients are usually characterized by high invasiveness and high metastatic rate.Many studies have shown that even after early diagnosis and intensive treatment,young breast cancer women are more likely to relapse and die than older women,so more attention needs to be paid to the prognosis of young women with breast cancer.At present,it is crucial to find stable and effective non-invasive biomarkers to accurately diagnose young breast cancer.MicroRNA is an endogenous,non-coding,small RNA that has been shown to be involved in a variety of cell?lar processes,including growth,survival,and apoptosis.The abnormal expression of microRNA is related to the pathogenesis of various cancers.At present,the potential diagnostic value of microRNA as a biomarker for breast cancer has become a hot spot.The current evidence-based medical diagnosis and treatment is mainly derived from the studies of middle-aged and elderly women,which is not enough to accurately assess the prognosis of young breast cancer.Therefore,for young breast cancer patients,new microRNA biomarkers are urgently needed to predict the prognosis,and develop a treatment strategy.Our group obtained samples of breast cancer patients of different ages from the First Affiliated Hospital of Fujian Medical University,and screened out miR-1908-3p,which is highly expressed by young breast cancer,through miRNAs gene chip technology.PCR technology was used to verify the expression characteristics of miR-1908-3p in different invasive breast cancer cells.In addition,tissue and serum samples(including young and old patients)of breast cancer patients werecollected from the hospital,and the expression level of differentially expressed miR-1908-3p in the young breast cancer was tested.The statistical res?lts were as follows: Compared with normal breast cell MCF10 a,miR-1908-3p was highly expressed in breast cancer cells,and the expression level of miR-1908-3p increased with the increase of invasiveness.High expression of miR-1908-3p in breast cancer tissues and serum was obtained from tissue and serum samples of total breast cancer patients.From the analysis of age information of breast cancer patients,miR-1908-3p was highly expressed in young breast cancer samples compared with tissues and serum of elderly breast cancer patients.This res?lt indicates that miR-1908-3p may be a biomarker for young breast cancer for clinical diagnosis.The clinical significance of miR-1908-3p in the diagnosis and prognosis of young breast cancer patients was clarified,but it is not clear how miR-1908-3p reg?lates the process and mechanism of breast cancer.To investigate the mechanism of action of miR-1908-3p in breast cancer,we transfected breast cancer cells with miR-1908-3p mimics,miR-1908-3p inhibitors,and cell proliferation,migration and invasion experiments.The changes in proliferation,migration and invasion ability of breast cancer cells were observed in the case of altered expression of miR-1908-3p.The res?lts showed that miR-1908-3p enhanced the proliferation,migration and invasion of MCF7 and MDA-MB-231 cells.We conclude that miR-1908-3p may achieve cancer-promoting effects in breast cancer by promoting the metastasis of breast cancer cells.The effect of miR-1908-3p on the biological behavior of cells was exated by cell function experiments,and further understanding of the molec?lar mechanism of miR-1908-3p reg?lation of breast cancer is needed.To clarify the reg?latory mechanisms,the focus is on finding proteins that are reg?lated by miR-1908-3p and that reg?late tumor cell migration.Therefore,we used Target Scan and miRanda databases to screen for target genes of miR-1908-3p.According to the biological role of miR-1908-3p in breast cancer,CRABP2 was finally selected as a candidate target gene for miR-1908-3p,which is reported to be closely related to tumor metastasis,and it is also one of the genes closely related to the pathophysiological process of breast cancer.The effects of miR-1908-3p on the mRNA and protein levels of CRABP2 were verified by real-time PCR and Westernblot.It was concluded that changes in the expression level of miR-1908-3p affect the mRNA expression level and protein expression level of CRABP2.The res?lts suggest that miR-1908-3p may promote the metastasis of breast cancer by reg?lating the expression of CRABP2,and further deepening the experiment,which remains to be explored to elucidate the mechanism of miR-1908-3p promoting cancer.
