Newcastle Disease Virus(NDV)belongs to the paramyxaviridae family of avian viruses,which can specifically infect and kill tumor cells.Through the clinical experiment verification,NDV is very weak to the human body's harm,may clear the virus quickly through own adjustment,therefore NDV may regard as one kind of very safe oncolysis virus.NDV can not only directly induce tumor cell death,but also induce the human immune system to produce various cytokines or affect the relevant immunogenicity of tumor cells to kill tumor cells.Due to the poor efficacy of traditional cancer therapies and the characteristics of oncoviruses that can kill tumor cells specifically,so NDV has become a very hot field for cancer treatment.Liver cancer is considered as the sixth most frequent malignant tumor in the world and has the characteristics of high degree of malignancy,rapid disease development and poor recovery after surgery,so liver cancer patients present a high mortality rate.Currently,liver cancer patients are mainly treated by the traditional method of surgical resection,at the same time with some auxiliary means,such as radiotherapy,chemical medication,However,the efficacy of these therapies is not satisfactory,and there are even serious side effects.Therefore,it is urgent to develop new treatment methods for liver cancer.At this time,oncovirus therapy for tumor began to appear in the researchers' field of vision,but it is still very difficult to promote NDV therapy for tumor to clinical application,which is also a problem to be solved at present.This paper mainly explored the mechanism of USP13 inhibiting NDV-induced apoptosis of liver cancer cells.First,two different liver cancer cell lines Huh7 and HLCZ01 were respectively infected with NDV.We found that both mRNA and protein of USP13 showed a downregulation trend,suggesting that USP13 plays an important role in NDV-induced apoptosis of liver cancer cells.The overexpression and silencing plasmids of USP13 were constructed,respectively,and it was found that overexpression of USP13 in the two liver cancer cell lines inhibited NDV-induced apoptosis of liver cancer cells,and conversely,knockdown of USP13 level promoted NDV-induced apoptosis of liver cancer cells.Finally,in the process of exploring the mechanism,we found that USP13 did not affect the replication of NDV in cells,but enhanced the anti-apoptotic molecule Bcl-2,thereby inhibiting the apoptosis of liver cancer cells.Based on those,we believe that USP13 may inhibitNDV-induced apoptosis of liver cancer cells by enhancing Bcl-2 expression.This sduty enriches the mechanism of NDV oncolysis in tumor cells and provides a new theoretical basis for the treatment of liver cancer by oncoviruses.
