Objective: Study the expression difference of hsa?circ?0092306 in colorectal cancer and normal adjacent tissues,compare its expression level in blood of healthy people and colorectal cancer patients,and explore its clinical significance.Methods:1.Trizol method was used to extract total RNA from 50 colorectal cancer tissues,corresponding normal intestinal tissues,50 colorectal cancer patients plasma and 50 healthy people plasma.2.RNA integrity was identified by agarose gel electrophoresis.3.Reverse transcription was performed using D7168 Beyo RT II c DNA first strand synthesis kit.4.CDNA was synthesized using D7262 Beyo Fast SYBR Green q PCR Mix.5.Agarose gel electrophoresis verified the molecular weight of the target product and identified it with the primer dimer.6.We compared the expression level of hsa?circ?0092306 in different samples and clarified its relationship with clinicopathological factors.Results: The expression level of hsa?circ?0092306 in colorectal cancer was down-regulated compared with that in normal intestinal tissue.Compared with the plasma of healthy people,the expression level of hsa?circ?0092306 in the plasma of colorectal cancer patients was decreased,and the decreased expression level was significantly correlated with the tumor size.Conclusions:1.Hsa?circ?0092306 is down-regulated in colorectal cancer tissues.2.The expression level of hsa?circ?0092306 in the plasma of colorectal cancer patients is decreased,and hsa?circ?0092306 is a new promising biomarker for colorectal cancer.
