Objective:This study was aimed to compare the efficacy and toxicity of gemcitabine combined with S-1 and gemcitabine combined with nab-paclitaxel as first-line treatment for advanced pancreatic cancerMethods:We conducted a retrospective single-center cohort study of 42 patients with advanced pancreatic cancer who received either GS(gemcitabine and S-1,21)or AG(gemcitabine and nab-paclitaxel,21)chemotherapy.The objective effective rate(ORR)?disease control rate(DCR)?median progression-free survival(PFS)?overall survival(OS)and adverse reactions of patients were analyzedResults:(1)There was no significant difference between S-1 plus gemcitabine group and nab-paclitaxel plus gemcitabine group in objective response rate(9.5%vs 23.8%,P>0.05),disease control rate(71.4%vs 85.7%,P>0.05),median progression-free survival(4.05 months vs 4.90 months,P>0.05)and median overall survival(11.48 months vs 9.05 months,P>0.05).(2)The adverse reactions of the blood system in both groups were high,76.2%and 80.9%,respectively,of which grade 3/4 hematological adverse reactions accounted for about half of the total population(42.9%and 47.6%,respectively).The incidence of non-blood system adverse reactions was low,mainly including elevated transaminase,fatigue,nausea,vomiting,diarrhea,rash,etc.,and the difference between the two groups was not statistically significant(P>0.05)Conclusions:In this retrospective study,it was found that first-line treatment with S-1 ombined with gemcitabine in advanced pancreatic cancer had similar effect and adverse eactions compared with the regimen of albumin paclitaxel combined with gemcitabine.
