| ObjectiveEpithelial ovarian cancer is a common malignant tumor of the female reproductive system,due to the unknown etiology and pathogenesis,the five-year survival rate of advanced patients is only 40%,and the mortality rate is the highest among gynecological malignancies.Recent studies have shown that changes of the microorganism in the human microenvironment may be related to the occurrence of human tumors.Current research finds that the female reproductive tract has a unique microorganism structure,and the pelvic cavity is the microenvironment in which the ovary exists,so it has greater research value and clinical significance.This project aims to explore the characteristics of pelvic microorganism in patients with epithelial ovarian cancer,clarify the correlation between pelvic microorganism and epithelial ovarian cancer,and provide effective ideas and values for the diagnosis and prognosis of ovarian cancer.MethodsThe study subjects were 50 surgical patients from Sir Run Run Shaw Hospital and Zhejiang Cancer Hospital,divided into epithelial ovarian cancer group,benign ovarian tumor group,and uterine fibroids group(control group),50 Douglas nest fluids were collected.Divided into groups based on clinical characteristics such as histological type,pathological grade,and lymph node metastasis.The 16 Sr RNA high-throughput geneticsequencing was used to determine the type and abundance of pelvic microorganism,filter the differential microorganism,construct the diagnostic model,analysis function of the microorganism,and filter related pathways.Results(1)Analysis of the 16 Sr RNA high-throughput genetic sequencing shows that patient with epithelial ovarian cancer has a unique microorganism structure,which is significantly different from the fibroid group,but it is not significant for benign ovarian tumor group.(2)Based on the observed OTUs,it shows that the species richness of the ovarian cancer group was significantly higher than benign ovarian tumor group and uterine fibroids group.(3)At the level of phylum,among the ovarian cancer microorganism,Bacteroidetes is the dominant microorganism,which has high microorganism abundance.At the level of genus,Parabacteroides and Prevotella have higher relative abundance.(4)The diagnostic model constructed by the differential microorganism between groups has significant diagnostic predictive value between ovarian cancer and the control group.(5)We found that,compared with the control group(uterine fibroids group),the ovarian cancer group was significantly enriched and expressed in metabolic pathways such as fatty acid metabolism,sphingosyllipid conversion,mucopolysaccharide biosynthesis and glycosylation.The ovarian benign tumor group was significantly enriched in fatty acid metabolism,polysaccharide degradation,riboflavin metabolism,and steroid metabolism.ConclusionsWe found that the pelvic microorganism of epithelial ovarian cancer has a unique structure,which is significantly different from the control group.It has a relatively stable differential microorganism structure.There is relatively stable differential microorganism structure.It also has significant functional enrichment differences with the blank control group and the benign ovarian tumor group in terms of microorganism function.This suggests that changes in the pelvic microenvironment of ovarian cancer and the occurrence of epithelial ovarian cancer may have a significant correlation,providing potential value for the occurrence,progression,early diagnosis and treatment of ovarian cancer. |
