Objective To study the relationship between AGT gene(rs699)VEGF gene(rs1570360 ? rs3025039 ? rs833061)single nucleotide polymorphisms and susceptibility to bladder cancer and kidney cancer.Method A 1: 1 matched case-control study was used,the project from January2018 to October 2019,patients were derived from kidney cancer and bladder cancer patients admitted to the urology department of the Affiliated Hospital of Inner Mongolia Medical University and Inner Mongolia Autonomous Region People's Hospital,the control were came from the hospital's physical examination center;the matching principle between controls and cases was that the difference between the same sex and age was not more than 3 years old.Face-to-face questionnaires were collected after the informed consent was obtained from the selected study population to collect baseline data.The genotypes of rs699,rs1570360,rs3025039,and rs833061 were detected by iMDR method.ELISA detection kit was used to determine the expression of VEGF in serum.Real-time quantitative PCR was used to determine the relative expression of VEGF mRNA cancerous and paracarcinomatous tissues.Use Epidata3.1 to enter the questionnaire,IBM SPSS25.00 was used for statistical analysis.Quantitative data follow the normal distribution was discribed“mean and standard”,the data was not obey the normal distribution and was described by "median(interquartile distance)",quantitative data obey normal distribution and apply t test,the data did not obey the normal distribution and the rank sum test was used.Categorical variable was discribed by composition ratio,categorical variable and Hardy-Weinberg equilibrium test was compared by chi-squaretest.Binary logistic regression analysis was used to calculate the OR value and 95% CI of the genetic model.Multivariate conditional logistic regression was used to analyze the relationship between daily living habits and eating habits and kidney and bladder cancer.The generalized multifactor dimensionality reduction method was used to analyze the gene-gene and gene-environment interactions,and the dominant models related to disease occurrence were screened,binary logistic regression analysis was used to calculate the OR value and 95% CI of the dominant model.Result 1.Univariate analysis showed that compared with the control group,the bladder cancer case group had differences in BMI,education level,smoking behavior,drinking behavior,dietary taste,fruit consumption,diet pattern,cooking oil,drinking water type,labor intensity,a full bladder,there were statistically significant differences in pesticide exposure history and factory work history(P <0.05);there were no statistically significant differences in hypertension,diabetes,frequency of diet,dairy products,water consumption,and air fresheners(P> 0.05).There were statistically significant differences between the kidney cancer case group and the control group in terms of education level,drinking behavior,frequency of diet,dietary taste,fruit consumption,dietary pattern,cooking oil,type of drinking water,labor intensity,a full bladder,and factory work history(P <0.05);there were no significant differences in BMI,hypertension,diabetes,smoking behavior,dairy products,water consumption,air fresheners,pesticide exposure history,and hair dyes(P> 0.05).2.Logistic regression analysis results of multivariate conditions showed that drinking non-pure water,a full bladder,factory work history,and pesticide exposure history were risk factors for bladder cancer(OR values were 2.088,1.600,3.776,1.803,P <0.05),Drinking behavior and factory work history were risk factors for renal cancer(OR values were 2.545,3.387,P <0.05).3.This study showed that the rs833061 CC vs.TT and CC vs.CT + TT model renal cancer group was statistically significant compared with the control group(OR= 0.347,0.363),suggesting that the C allele is kidney cancer relative to the T allele protective factors.4.GMDR analysis showed that rs833061,factory work history,a full bladder,and drinking non-pure water(OR = 2.108,P = 0.001)were the dominant models of bladder cancer interaction.rs3025039,rs699,rs833061,drinking and factory work history(OR = 1.540,P <0.001),rs1570360,rs3025039,rs699,rs833061,drinking and factory work history are the superior models of renal cancer interactions(OR = 1.390,P = 0.001).5.The expression levels of serum VEGF in bladder cancer and kidney cancer groups were significantly higher than those in the control group(P <0.001).6.The difference of VEGF mRNA between kidney cancer cancerous and paracarcinomatous tissues was not statistically significant(P = 0.445).7.The rs699 locus genotype was related to the pathological grade and clinical stage of bladder cancer(P values were 0.049 and 0.033),and the rs1570360 genotype was related to the pathological grade of renal cancer(P = 0.020).Conclusion 1.The risk factors for bladder cancer are drinking unpurified water,a full bladder,history of factory work,and history of pesticide exposure;risk factors for kidney cancer are drinking behavior and history of factory work.2.This study showed that the rs833061 CC vs.TT and CC vs.CT + TT model renal cancer group was statistically significant compared with the control group(OR= 0.347,0.363),suggesting that the C allele is kidney cancer relative to the T allele protective factors.3.rs833061,factory work history,a full bladder and drinking non-pure water are the dominant models of bladder cancer interactions.rs3025039,rs699,rs833061,drinking behavior and factory work history,rs1570360,rs3025039,rs699,rs833061,drinking behavior and factory work history are the dominant models of renal cancer interaction.4.The expression of serum VEGF in bladder cancer and kidney cancer groups was significantly higher than that in control group.5.There is no difference in the expression of VEGF mRNA between kidney cancer cancerous and paracarcinomatous tissues.6.The rs699 locus genotype is related to the pathological grade and clinical stage of bladder cancer,and the rs1570360 genotype is related to the pathological grade of renal cancer.
