Objective:Explore the expression of protein arginine methyltransferase 5(PRMT5)in gastric cancer and its clinical significance.Methods:Eighty-three patients with gastric cancer were selected from the affiliated Hospital of Inner Mongolia Medical University from January 2011 to January 2013,including66 males and 17 females,with an average age of 57.17±8.88 years old.All participants met selection criteria and exclusion criteria.The tissue wax blocks of 83 patients with gastric cancer were randomly selected,and the corresponding adjacent normal tissues(the surgical margin is more than 5cm from the tumor)were selected as the control group.Immunohistochemistry(IHC)method was used to detect the expression of PRMT5 in gastric cancer tissues and adjacent normal tissues.At the same time,IHC method was used to detect the expression of Antigen Ki67(Ki67)in the same gastric cancer tissue.The relationship between the expression of PRMT5 and the clinic pathological characteristics of gastric cancer,the expression of Ki67 and prognosis were analyzed using statistical methods.All data were statistically analyzed using SPSS 20.0(SPSS Inc.,Chicago,USA)software.Two-tailed tests of significance were reported,with P<0.05 considered statistically significant.Results:1.IHC assay showed that PRMT5 was expressed in both nucleus and cytoplasm,mainly in nucleus in gastric cancer tissues and in cytoplasm in normal tissues adjacent to cancer.The high expression rate of PRMT5 in gastric cancer was 60.2%(50/83),and the high expression rate in normal tissues adjacent to cancer was 18.1%(15/83).The high expression rate of PRMT5 in gastric cancer tissues was higher than that in normal tissues adjacent to cancer,and the difference was statistically significant(P<0.001).2.The ratio of poorly differentiated and undifferentiated,infiltrated depth T3 and T4,lymph node metastasis and clinical stage III and IV in PRMT5 high expression group was higher than that in low expression group,the difference was statistically significant(P<0.05);There was no significant difference between PRMT5 high expression group and low expression group in age,gender,and expression ratio of preoperative tumor marker Carbohydrate Antigen199(CA199)and Carcinoembryonic Antigen(CEA)(P>0.05).3.IHC assay showed that the expression of Ki67 was located in the nucleus.The common high expression of Ki67 and PRMT5 in gastric cancer tissues was 38 cases,the high expression rate was 45.8%,the common low expression rate was 25 cases,and the low expression rate was 30.1%.The results of Spearman correlation analysis showed that there was a positive correlation between PRMT5 and Ki67 expression in gastric cancer tissues(r_s=0.507,P<0.001).4.The overall survival(OS)of patients with high expression of PRMT5 was significantly lower than that of patients with low expression,and the difference was statistically significant(P<0.05).Univariate analysis showed that the degree of differentiation,depth of invasion,lymph node metastasis,clinical stage,preoperative tumor markers CA199,Ki67 and PRMT5 expression had influence on the prognosis of gastric cancer.Multivariate results showed that PRMT5 expression,Ki67 expression,and clinical stage were independent prognostic factors of gastric cancer.Conclusions:1.PRMT5 is highly expressed in gastric cancer tissues,and its high expression is related to the adverse clinical and pathological characteristics of gastric cancer,indicating that PRMT5 may be a cancer factor and participate in the growth,proliferation,invasion and metastasis of gastric cancer.2.There is a significant positive correlation between the expression of PRMT5 and Ki67in gastric cancer,indicating that PRMT5 may be involved in the growth and proliferation of gastric cancer,but the specific mechanism remains to be further elucidated.3.High expression of PRMT5 is a risk factor for the prognosis of gastric cancer,which is helpful to evaluate the metastasis,invasive ability and prognosis of gastric cancer.
