Background: Lung cancer is one of the common malignant tumors that seriously threaten the physical and mental health of human beings.It is mainly divided into Small Cell Lung Cancer(SCLC)and Non-Small Cell Lung Cancer(NSCLC),Non-small cell lung cancer is most common.Due to the lack of effective diagnostic and treatment methods,the morbidity and mortality worldwide have been high and showing an increasing trend.Therefore,in-depth study of the molecular mechanism of its occurrence and development is urgently needed to provide new diagnostic and treatment of non-small cell lung cancer Ideas.Long non-coding RNAs(lnc RNAs)LINC01614 is located in the long arm of chromosome 2.The role of this gene in the development of non-small cell lung cancer has not yet been determined.Objective: 1.To detect the expression of LINC01614 in non-small cell lung cancer tissues and non-small cell lung cancer cell lines.2.Decrease the expression level of LINC01614 in non-small cell lung cancer cell lines,and explore the effects of this gene on proliferation,invasion and metastasis,and apoptosis of non-small cell lung cancer cell lines.Method:1.To explore the expression of LINC01614 in non-small cell lung cancer.At the tissue level,q RT-PCR was used to detect 10 non-small cell lung cancer tissues and adjacent normal tissues.At the cellular level,q RT-PCR was used to detect the human non-small cell lung cancer cell line A549 and the human normal bronchial epithelial cell line BEAS-2B.2.Construction of LINC01614-knockout A549 cell model using CRISPR-cas9 technology.3.CCK-8 detected the proliferation level of A549 cell line after knocking out LINC01614.4.Scratch injury experiment to study the invasion ability of A549 cell line after knocking out LINC01614.Results:1.The expression level of LINC01614 in non-small cell lung cancer tissues was significantly higher than that in normal adjacent tissues(p <0.05),the difference was statistically significant.2.The expression level of LINC01614 in human non-small cell lung cancer cell line A549 was significantly higher than that of human normal bronchial epithelial cell line BEAS-2B(p <0.05),the difference was statistically significant.3.CCK-8 experimental results show that compared with the control group,the cell proliferation of the experimental group was inhibited(p <0.05),and the difference was statistically significant.4.Scratch damage experiment results show that compared with the control group,the cell invasion ability of the experimental group is inhibited(p <0.05),and the difference was statistically significant.5.Flow cytometry results showed that compared with the control group,the apoptosis level of the experimental group was enhanced(p <0.05),and the difference was statistically significant.Conclusion:1.LINC01614 is highly expressed in non-small cell lung cancer tissues and non-small cell lung cancer cell lines.2.Decreasing the expression level of LINC01614 in non-small cell lung cancer cell lines can inhibit cell proliferation,migration ability.
