Objective:Through the observation of the effect of Gei Wang-9 on the general activity,biochemistry,and pathology tempreture,mortality,survival time of rats with acute liver failure induced by D-Gal N,to illustrate preventing and curing effects of Gei Wang-9 on acute liver failure Methods:72 SPF,male,weighting 200± 20 SD rats devide into 6 groups:normal control group,model group,positive control group,low-dose group,medium-dose group and high-dose group,randomly.Firstly,The rats in both normal control group and model group were given 0.5% CMC-Na solution,positive control group were given Compound Glycyrrhizen tablets 0.45g/kg and Gei Wang-9 low,medium,high groups were given 0.4/g/kg,0.8g/kg,1.6g/kg in 0.5%CMC-Na solution by intragastric administration 20ml/kg 10 days,continuely.Then at the 11 th day,2 hour after the Rats in each groups had intragastric administrated the drugs,To induce the Acute liver disease model by D-Gal N 600mg/kg intraperitoneal injection.lastly,To investigate the general activity 48 hours and then isolated the liver tissue,blood serum,blood plasma to observe the histopathological changes and test the ALT,AST,T-Bil,D-Bil,TP,ALB,CHE level by automatic biochemical analysis instrument;test the time of PT,APTT by Automatic Plasma Analyzer and observe the color,scar and shape of the liver and fixed,dehydration,paraffin embedding,HE staining to observe the pathology.The 72 rats were divided into six groups like the method of part 1.After adaptive feeding for 3 days,induced acute liver failure by D-Gal N according to part 1.2 hours later,Gei Wang-9 and positive control groups were administrated the medicine as part 1.observe the activity,measured the body temperature every 2hours and record survival time and sum up with the mortality during 96 h.Results:1.General activity : ?1 blank group good glossiness,move freely,response sensitively,normal ?diet,defecation and weight growth.?2 model group slow in response,listless,drowsy,fluffy messy ? ?fur was soaked with sweat and itchy.48 hours later,drowsiness,pain response disappearing,deep yellow urine,limb or convulsion,mania,abdominal distention and bleeding at the end of oral or limb.positive control group and the Gei Wang-9 groups: less of the above symptoms than model group: no such symptoms as the whole body fur was fluffy and sweat soaked,The urine color in the Gei Wang-9 group was lighter than that in the model group.2.Liver biochemistry :(1)compared with the blank group,ALT and AST of the model group were significantly increased(P < 0.01,P < 0.01).Compared with the model group,ALT of rats in the low,medium dose Gei Wang-9 group was reduced(P < 0.01,P < 0.01,P < 0.05).and the high dose group was no statistically sidnificant(P > 0.05).Compared with the model group,the ASTvalue of each treatment group was decreased,but there was no significant difference(P > 0.05).(2)Compared with the normal group,the T-Bil and D-Bil contents of the model group were increased,value of eachtreatment group was decreased,but there was no significant difference(P > 0.05).(2)Compared with the normal group,the T-Bil and D-Bil contents of the model group were increased,the difference was statistically significant(P < 0.05,P < 0.05).The T-Bil values of the positive control group and the low dose group of Gei Wang-9 were lower than the model group,but no statistical significance(P > 0.05).The T-Bil contents of the high and medium dose groups of Gei Wang-9 were significantly lower than the model group,(P < 0.05);There was no significant difference between the control group and the model group(P > 0.05).(3)Compared with the blank control group,the TP ALB CHE of the model group were significantly reduced(P < 0.05),but there was no significant difference between the positive control group and the high,medium and low dose group(P > 0.05).CHE value of the positive control group was significantly lower(P < 0.05).There was no statistical significance in the decrease with CHE value of the high,medium and low dose group of Gei Wang-9(P > 0.05).3.Effects of coagulation: compared with the normal group,the PT,APTT model group was significantly longer(P < 0.05).Compared with the model group,the positive control group and the high,medium and low dose group of Gei Wang-9 was not significantly shorter(P > 0.05).4.Histopathological observation:(1)Blank group:the liver is ruddy and normal in size,without congestion and necrosis spots;liver cords is clear and complete,the shape and size of stem cells is normal,the nucleus is complete and full,there is no necrosis and inflammatory cells in the portal area.(2)Model group: the liver tissue structure is disordered,cord is not clear,swollen and deformed,most of the diffuse moderate to severe edema,occasionally with fatty degeneration the infiltration of plasmacytes and monocytes can be seen occasionally.there are diffuse large-scale and bridging necrosis of cells around the portal area of liver tissue and multiple nuclear pyknosis,dissolution and fragmentati.(3)Positive control group: liver tissue structure was disorder,cord was not clear,severe edema of liver cells was less than that in model group,infiltration of neutrophils and lymphocytes was reduced,no large-scale necrosis was found in portal area and liver lobe,bridging necrosis was less than that in model group,The histopathology was lower than that of model group,but the difference was not statistically significant(P > 0.05).(4)Gei Wang-9 groups : the arrangement of the liver plate,liver cord not improve significantly,steatosis was alleviated,infiltration of inflammatory cells around the portal area was alleviated,necrosis areas in the middle and high dose groups were significantly improved,The difference compared with modle group of pathological high,middle and low dose groups was statistically significant(P < 0.05).5.Body temperature: there was no significant difference between the basic body temperature groups(P < 0.05).The body temperature of each group changed regularly two hours after themodel.Low dose Gei Wang-9 played a role in lowering the body temperature caused by D-Gal N.6.General situation:(1)blank group good glossiness,move freely,response sensitively,normal eating,normal defecation and normal weight growth.(2)modelgroup: 12-24 hours,activities were reduced,drowsiness and respiration was increased;24-48 hours,drowsiness rats were increased,fluffy,body fur was soaked with sweat,48 hours later,pain response become slow or disappearing,yellow urine,convulsions of limbs or the whole body,mania,abdominal distention,bleeding at the end of the oral limbs,and nose bleed to asphyxia.(3)positive control group: the above symptoms less severe,and there were no symptoms such as fluffy,sweat soaking,itching,etc.bleeding in the mouth,nose and extremities resulted in asphyxia and death.Gei Wang-9 group: without obvious skin,fluffy sweat,pruritus,drowsiness and mental malaise.Most rats suffered from bleeding in the mouth,nose and extremities,resulting in blood loss and asphyxia.7.Effect of survival time and mortality :compared with blank groupsurvival time was decreased and mortality was increased in model group(P < 0.05).There was no significant difference in the survival time and mortality of control group and Gei Wang-9 high,medium,low dose group.Conclusion:1.The high dose of Gei Wang-9 can reduce the serum ALT,hepotocyte damage and necrosis.2.Gei Wang-9 high middle dose can decrease the level of serum T-Bil,D-Bil,as well as improve the symphtoms of billirabin thesaurismosis like yellow urine and fur.3.Every dose of Gei Wang-9 can not positive effect on blood coagulation when liver desease induced by D-Gal N.4.Every dose of Gei Wang-9 have no effect on TP compose function of liver and hepatocyte aquatic evidently.5.Gei Wang-9can release the symphtoms of acute liver failure of rats induced by D-Gal N,as sleeping,skin and urine yellow and dry stool.In summery,Gei Wang-9 can protect liver function from jaundice,protein components,and hepato pathology like neccerosis.However,there is no effect on blood coagulation induced by liver damage.so it is efficient that use Gei Wang-9 with hemostatic drugs in clinic tratment.
