Objective: The expression of the SphK-S1P-S1 PR signaling pathway related proteins(SphK1,SphK2,S1PR1,and S1PR2)in adult patients with Diffuse Large B-cell Lymphomas(DLBCL)was detected.The relationship between protein expression levels and clinical-pathological indicators were also further analyzed,including protein expression correlations and the evaluation of therapeutic effects.The objectives of this study were to find potential therapeutic targets for DLBCL.Methods:Immunohistochemistry(IHC)was used to evaluate the expression levels of SphK1,SphK2,S1PR1,and S1PR2 in paraffin lymphaden tissues.Samples were collected from 28 DLBCL patients and 10 patients with non-malignant lymph node disease.Results: 1.The IHC data showed that there was a strong correlation in protein expressions between two types of patients(P < 0.05).2.Fisher's exact tests suggested that among clinical pathological indicators,the SphK1 protein expression was associated with IPI score group;the S1PR1 expression was associated with age classification,IPI score and Bcl-2 expression groups;and the S1PR2 expression was associated with gender,ANN ARBOR stage,IPI score and LDH level groups(P < 0.05).3.The protein expressions between SphK1-S1PR1 and SphK1-S1PR2 in tissues of DLBCL patients were statistically correlated(P < 0.05).4.The short-term remission rate of groups with expressed SphK1,SphK2,S1PR1 protein were significantly lower than the non-expressed group,and an opposite relationship was observed for S1PR2 protein.The differences between the rates above were not statistically significant due to the small sample size.The sample size should be expanded for further verification in the future.Conclusions:Both SphK1,SphK2 and S1PR1 protein expressions and the removal of S1PR2 protein are strongly correlated to the pathogenesis of DLBCL.The increasing positive expression rates of SphK1 and S1PR1,and the decreasing positive expression rate of S1PR2 are reliable indicators of DLBCL patients with poor prognosis.Detecting the expression of the SphK-S1P-S1 PR signaling pathway related proteins may be an effective tool for the accurate diagnosis and prognosis judgement of DLBCL patients.Further analyse of the SphK-S1P-S1 PR signaling pathway is expected to provide new potential targets for DLBCL treatment.
