Objectives To observe the influence of Shuangqing PingHua decoction(SPHD)on biochemical indicators related to glucose metabolism,pancreatic tissues histopathological changes and the expression levels of phosphoinositide 3 kinase(PI3K),protein kinase B(AKT),phospho-AKT(p-AKT),forkhead box protein O1(FOXO1)and phospho-FOXO1(p-FOXO1)proteins in rats with metabolic syndrome(MS).To explore and the possible mechanism on rats with glucose metabolism and MS.Methods The male SD rats were randomly divided into two groups,54 in metabolic syndrome damge group(MSD group),6 in normal control group(NC group).The MSD group was fed with high fat diet combined with 10% fructose water.The NC group was fed with general diet and water until the end of the experiment.The MSD group was treated with L-NAME 20mg/(Kg?d)intragastric administration and streptozotocin(STZ)25mg/Kg intraperitoneal injection to build the model of MS.The body weight,body length,abdominal circumference,blood pressure and blood sugar were measured regularly in the modeling stage,and the modeling situation was evaluated in the 14 th week.The rats with MS were randomly divided into 5 groups-MSD group,Metformin group(MF group),SPHD groups with low-lose(SPHD-L),mid-lose(SPHD-M),high-lose(SPHD-H).The total amount of daily food intake would be devided into two portions,which was gave to rats regularly in the morning and evening.They were respectively fed with high fat feed,high fat feed with MF or low-lose SPHD or mid-lose SPHD or high-lose SPHD.The sugar tolerance were tested regularly in the treatment stage.At the 8th weekend,the abdominal aorta was collected to detect the level of fasting blood glucose(FBG),glycosylated serum protein(GSP),hemoglobin A1c(HbALc),superoxide dismutase(SOD)and malonaldehyde(MDA).The level of insulin(INS)and C-Peptide(C-P)were detected by the way of ELISA.The homeostasis model assessment-insulin resistance(HOMA-IR)and insulin sensitivity index(ISI)were calculated.The expression of PI3 K,AKT,FOXO1,p-FOXO1 protein in pancreatic tissues was detected by the way of Western bolt.The pathological condition was observed in pancreatic tissue with HE staining.Results 1 In the stage of building models,the NC group rats were generally in good condition,while the the MSD group rats were in poor state.Compared with NC group,the body weight,abdominal circumference,Lee's index,blood pressure,FBG and(area under curve)AUC was increased(P<0.05,P<0.01)in the end of 14 th weekend.2 The 8th week after treatment:the state of MF,SPHD-M and SPHD-H group slowly became better than the MSD group.The results of sugar tolerance showed that the level of AUC and blood glucose at every time point in the MSD group were significantly higher than NC group(P<0.01),while it was lower in the MF and SPHD-H group than the group of MSD(P<0.01).The level of AUC and the blood glucose at the time of 0min and 120 min were decreased in the SPHD-M group compared with MSD group(P<0.05,P<0.01).In the MF group,the level of AUC and blood glucose at the time of 0min,30 min,120min was lower than SPHD-H group and the level of AUC and blood glucose at every time point was decreased compared with SPHD-M group(P<0.01).Compared with SPHD-M group,the level of AUC and blood glucose at every time point in the SPHD-H group were decreased(P<0.01).Compared with the MSD group,the level of FBG,GSP,INS,HbALc,HOMA-IR,MDA were decreased in MF and SPHD-H group(P<0.01),and the C-P,ISI,SOD were increased(P<0.01),and the level of MDA,FBG,HOMA-IR were decreased and the ISI was increased in the group of SPHD-M(P<0.05,P<0.01).In the group of MF,the level of FBG was decreased and the ISI was improved compared with SPHD-H group(P<0.05),and the level of MDA,FBG,HOMA-IR were decreased and the ISI was improved compared with the SPHD-M group(P<0.01),while the level of GSP,HbALc,INS,C-P,HOMA-IR,SOD and MDA had no significantly difference compared with the SPHD-H group(P>0.05).3.The HE staining of pancreatic showed that the langerhans' islet of the NC group had regular morphology and distribution,while the internal structure of the langerhans' islet was disorderedand the number of cells was decreased and there were some vacuolar degenerations and nuclear contractions in the MSD group.The distribution of insulin in Chinese medicine group and MF group were stronger than those in MSD group.4.The results of the expression of protein were as follows.There was no significant difference in the expression of PI3 K and AKT proteins among every group.Compared with NC group,the level of p-FOXO1 /FOXO1 was decreased in each group(P<0.01),and the level of p-AKT/AKT was also decreased in each group except the MF group.The level of p-FOXO1 /FOXO1 and p-AKT/AKT were improved in the group of MF and SPHD-H(P<0.01).There were no difference between MF group and SPHD-H group in the levels of p-FOXO1 /FOXO1 and p-AKT/AKT.Conclusions 1 SPHD can reduce the blood glucose level of MS rats and correct the disorder of glucose metabolism.2 SPHD can reduce the level of MDA and improve the vitality of SOD which can resist oxidative stress.Therefore,SPHD maybe delay the progress of MS glucose metabolism disorde through resisting oxidative stress which can protect islet cells and improve insulin resistance(IR).3 SPHD can increase the levels of p-AKT/AKT and p-FOXO1/FOXO1 in the pancreas,so that SPHD maybe regulate the disorder of glucose metabolism by increasing the phosphorylation of AKT and FOXO1 protein,regulating the signal channel of
