Objectives Observe the effects of marrow cavity rehydration on the survival rate of sepsis mice,the number of bone marrow cells and liver cells,the number and proportion of regulatory dendritic cells(CD11clowCD45RBhigh DCs)in bone marrow and liver,explore the relationship between CD11clowCD45 RBhigh DCs and the immune function of sepsis mice and the effect on the prognosis of sepsis mice.Methods 1 The effect of marrow cavity rehydration on the number of bone marrow cells and liver cells in sepsis mice and the survival rate of sepsis mice: 30 healthy male C57BL/6 mice(6?8w),weight 201g.These samples were randomly divided into 3 groups: control group(A0),25?L/g liquid resuscitation group(A1),50?L/g liquid resuscitation group(A2),there are 10 mice in each group.In group A0,only the marrow cavity rehydration model was established without liquid injection,followed by intraperitoneal injection of lethal bacteria.Group A1 and A2 were given sterile saline 25?L/g and 50?L/g through bone marrow cavity,respectively.Subsequently,a single intraperitoneal injection of lethal bacteria was used to create a sepsis model.At the end of the operation,put it back in the cage and drink water freely.Observe the survival rate of the mice after 7 days.30 healthy male C57BL/6 mice were randomly divided into three groups: A0,A1,and A2.The operation of the three groups was the same as the observation survival experiment described above.The numbers and proportions of CD11clowCD45 RBhigh DCs in mouse bone marrow cells and liver cells and bone marrow cells and liver cells in each group were observed at 0d,3d,5d and 7d.2 Observe the effects of CD11clowCD45 RBhigh DCs in bone marrow cells and liver cells and bone marrow cells and liver cells on the prognosis of sepsis mice: 90 healthy male C57BL/6 mice(6-8w),weight 201g.These samples were randomly divided into 9 groups: control group(C),the number of bone marrow cells reinfused 5106 group(B1),the number of bone marrow cells reinfused group 107(B2),the number of liver cells reinfused 5106 groups(L1),the number of liver cells reinfused 107 groups(L2),the number of CD11clowCD45 RBhighDCs reintroduced into bone marrow cells 5105 group(BD1),the number of CD11clowCD45 RBhigh DCs reintroduced into bone marrow cells 106 group(BD2).The number of CD11clowCD45 RBhigh DCs in bone liver cells was 5105(LD1),and the number of CD11clowCD45 RBhigh DCs in liver cells was 106 groups(LD2),there are 10 mice in each group.Group C was injected intraperitoneally with the same amount of sterile saline,and the remaining groups were intraperitoneally injected with equivalent sterile saline containing different cell numbers.Observe the survival rate of each group after sepsis mice in each group.Results 1 The effect of fluid replacement of the marrow cavity on the survival rate of sepsis mice: three groups of mice had poor drinking water,poor appetite,and increased eye corner secretions.The survival rate of mice in group A0 was 0%,the survival rate of mice in group A1 was 50%,and the survival rate of mice in group A2 was 70%.Compared with group A0,the survival rate of sepsis mice in A1 and A2 groups were significantly improved(P<0.05);compared with group A1,the survival rate of sepsis mice in groups A2 was significantly improved(P<0.05).2 Changes in the number of bone marrow cells and liver cells: compared with group A0,the number of bone marrow cells and liver cells in groups A1 and A2 increased(P<0.05),showing a dose-effect relationship.3 Changes in the number of CD11clowCD45 RBhigh DCs in bone marrow and liver: compared with group A0,the number of CD11clowCD45 RBhigh DCs in groups A1 and A2 increased significantly(P<0.05);compared with group A1,the number of CD11clowCD45 RBhigh DCs in group A2 increased significantly(P<0.05).4 Changes in the ratio of CD11clowCD45 RBhigh DCs to DCs in bone marrow and liver: compared with group A0,the proportion of CD11clowCD45 RBhigh DCs/DCs in groups A1 and A2 increased significantly(P<0.05);compared with group A1,the proportion of CD11clowCD45 RBhigh DCs/DCs in group A2 increased significantly(P<0.05).5 The effect of cell transfusion on the survival rate of sepsis mice: 1)Cell transfusion of bone marrow cells and liver cells can reduce the mortality of sepsis mice,but the difference is not statistically significant(P>0.05).2)CD11clowCD45RBhigh DCs in bone marrow reinfusion: compared with group C,the survival rate of sepsis mice in BD1 and BD2 groups were significantly improved(P<0.05);compared with BD1 group,the survival rate of sepsis mice in BD2 group was significantly improved(P<0.05).3)CD11clowCD45RBhigh DCs reinfused into liver: compared with group C,the survival rate of sepsis mice in LD1 and LD2 groups were significantly improved(P<0.05).Conclusions 1 Marrow cavity fluid can increase the number of bone marrow cells and liver cells in sepsis mice,and increase the survival rate of sepsis mice.2 Marrow cavity fluid can increase the number and proportion of CD11clowCD45 RBhigh DCs in bone marrow cells and liver cells of sepsis mice,and increase the survival rate of sepsis mice.3 The number of CD11clowCD45 RBhigh DCs can affects the immune function of sepsis mice,reverses the late immune paralysis of sepsis mice and improves the survival rate of sepsis mice.Figure 12;Table 2;Reference 157...
