Protective Effect Of Arctiin Combined With Ginkgo Biloba On Retina Of Diabetic Rats

Objectives To observe the effect of arctiin combined with ginkgo biloba extract on vascular endothelial growth factor(VEGF)and platelet-derived growth factor-B(PDGF-B)in the retina of diabetic rats,in order to explore the protective effect of arctiin combined with ginkgo biloba extract on the retina of diabetic rats.Methods Forty-five clean grade healthy male sprague-dawley(SD)rats(weight 200g~250g)were selected by random sampling.The conditions of the anterior segment and fundus of each rat were examined by fundoscope,and no obvious abnormalities were found.Nine rats were randomly selected from 45 rats as the normal group and the remaining 36 as the experimental group.The experimental group was intraperitoneally injected with 1% streptozotocin(STZ)diluent to establish a diabetic rat model.In building successful rats,which has 36 rats,randomly divided into 4 groups: the injury group(9 rats are given only drinking water by intragastric administration),the arctiin group(9 rats are given arctiin solution by intragastric administration),the ginkgo biloba group(9 rats are given ginkgo biloba extract solution by intragastric administration),the arctiin combined with ginkgo biloba group(9 rats were given arctiin and ginkgo biloba extract solution by intragastric administration),and 9 rats in the normal group were given drinking water by intragastric administration.Venous blood was collected from the tail of each group to monitor the changes of blood glucose and the anterior segment and fundus of rats were observed by slit-lamp ophthalmoscope.Rats in each group were sacrificed after 12 weeks of administration and their eyeballs were removed.HE staining was used to observe the retinal morphology of rats in each group,and the expression of VEGF and PDGF-B proteins in the retina of rats in each group was observed by immunohistochemistry.Results At the 12 th week of the experiment: 1 The body weight of rats in the injury group was lower than that in the normal group,and the difference was statistically significant(P<0.05);The body weight of rats in the treatment group(including the arctiin group,the ginkgo biloba group and the arctiin combined with ginkgo biloba group)was higher than that in the injury group,and the difference was statistically significant(P<0.05);The body weight of rats in the arctiin combined with ginkgo biloba group was higher than that of the single drug group(including the arctiin group and the ginkgo biloba group),and the difference was statistically significant(P<0.05).2 The blood sugar of rats in the injury group was higher than that in the normal group,and the difference was statistically significant(P<0.05);The blood sugar of rats in the treatment group was lower than that in the injury group,and the difference was statistically significant(P<0.05);The blood sugar of rats in the arctiin combined with ginkgo biloba group was lower than that of the single drug group,and the difference was statistically significant(P<0.05).3 The fundus lesions in the injured group were more serious than those in the normal group,and the difference was statistically significant(P<0.05).The fundus lesions in the treatment group were milder than those in the injury group,and the difference was statistically significant(P<0.05);The fundus lesions in the arctiin combined with ginkgo biloba group were milder than those in the single drug group,and the difference was statistically significant(P<0.05);4 HE staining results showed that the retinal layers of rats in the normal group had complete,clear structures and regular arrangement;In the injured group,the retinal tissue was highly edema,the number of ganglion cells was reduced,and the outer nuclear layer was disordered,loose and thin;In the arctiin group and the ginkgo biloba group,there was moderate edema in the retinal tissue,the number of ganglion cells was slightly decreased,and the outer nuclear layer was irregular;The structure of each layer of the retina in the arctiin combined with ginkgo biloba group was complete,clear and regular,and no obvious abnormal changes were observed.5 Immunohistochemical results showed that the expression of VEGF and PDGF-B in the retina of rats in the injured group was higher than that in the normal group,and the difference was statistically significant(P<0.05).The expression of VEGF and PDGF-B in the retina of rats in the treatment group was lower than that in the injured group,and the difference was statistically significant(P<0.05).The expression of VEGF and PDGF-B in the retina of the arctiin combined with ginkgo biloba group was lower than that of the single drug group,and the difference was statistically significant(P<0.05).Conclusions 1 Both arctiin and ginkgo biloba extract had protective effects on early retinal damage in diabetic rats.2 Arctiin combined with ginkgo biloba extract has a better protective effect on early retinal injury in diabetic rats.Figure 20;Table 7;Reference 161...
