Objective To evaluate the biocompatibility of m XG gel and establish adhesion model of cecum-abdominal wall injury in Kunming mice,and to comprehensively evaluate the effect of m XG gel combined with C5 a antibody in preventing abdominal adhesion.Methods 1 The biocompatibility of m XG gel was evaluated by CCK-8 assay,live/dead cell staining and 3D cell culture.2 The adhesion model of cecum-abdominal wall defect in kunming mice was made to evaluate the effect of m XG gel combined with C5 a antibody in preventing adhesion after abdominal surgery.48 Kunming mice were randomly divided into saline control group;C5a antibody group;m XG group and m XG/C5 a antibody group.Six animals were sacrificed at 1 week and 2 weeks after the operation.The degree of adhesion was visually scored by double-blind method.HE,VG,Masson and Sirius red staining were performed on the injured site for histopathological observation.The expression of TGF-?1 in the injured sites was observed by immunohistochemical staining.45 Kunming mice were randomly divided into normal group;C5a antibody group;operation group.Five animals in each of the three groups were sacrificed on the 1st,3rd and 5th day after the operation.The expression of IL-8 and neutrophils at the injury site were observed by immunohistochemical staining,and the expression of IL-6 and IL-8 in the serum was detected by Luminex technique.Results 1 The CCK-8 assay indicated that m XG gel had good biocompatibility.Live/dead cell staining indicated that m XG gel had no cytotoxicity.3D cell culture confirmed that m XG gel had good biocompatibility.2 At the 1st and 2nd week after the operation,the results of adhesion grading score,HE,VG,Masson,Sirius red staining and immunohistochemistry showed that m XG/C5 a antibody could effectively prevent the occurrence of adhesion,and reduce the inflammatory response while playing a role in physical isolation,thereby promoting the healing of wound tissue.The results of immunohistochemical staining and Luminex indicated that the intervention of C5 a antibody inhibited the excessive activation of complement and inhibited the inflammatory response,and the synergistic treatment could prevent the occurrence of abdominal adhesion.Conclusion 1 m XG gel has good biocompatibility.2 m XG thermosensitive hydrogel in combination with C5 a antibody can more effective in preventing the formation of postoperative abdominal adhesions.3 C5 a antibody can reduce the inflammatory response by reducing the content of inflammatory factors and inflammatory cells in the tissue and serum at the site of injury.Figure 16;Table 2;Reference 94.
