| Renal cell carcinoma(RCC)has continued to rise in recent years,ranking third in malignant tumors of the urinary system.More and more reseraches have found that long-chain non-coding RNAs(Lnc RNAs)plays an important role in the occurrence and development of tumors.It had been confirmed that long-chain non-coding RNA ZNF667-AS1 showed low expression and hypermethylation in cervical cancer and breast cancer respectively.ZNF667 was involved in the occurrence and development of hepatocellular carcinoma.ZNF667-AS1 and ZNF667 showed low expression and abnormal hypermethylation in esophageal cancer tissues and esophageal cancer cells.However,the role of ZNF667-AS1 in RCC has not been reported.The purpose of this reserach is to study and explore the possible role and mechanism of ZNF667-AS1 and ZNF667 in RCC,which could provide a theoretical basis for further research.The main contents of this reserach are as follows:Objective: To detect the methylation status of ZNF667-AS1 in RCC and corresponding normal tissues,and analyze the relationship with clinicopathological data of patients.To detect the expression levels of ZNF667-AS1 and ZNF667 in RCC and corresponding normal tissues,and analyze the relationship with clinicopathological data of patients.To explore the correlation between the expression of ZNF667-AS1 and ZNF667 in RCC tissues.Methods:1.Methylation specific polymerase chain reaction(MSP)method was applied to detect the methylation status of ZNF667-AS1 Cp G islands of various regions in 59 RCC and corresponding normal tissues.2.Real-time quantitative polymerase chain reaction detecting system(RT-q PCR)was applied to detect the expression levels of ZNF667-AS1 and ZNF667 in 59 RCC tissues and corresponding normal tissues.Results:1.Methylation status of ZNF667-AS1 Cp G islands of various regions in RCC and corresponding normal tissues: in RCC tissues,the methylation rates of ZNF667-AS1 Cp G islands of various regions were higher than those in corresponding normal tissues((67.8% vs.39.0%),(84.7% vs.16.9%),(72.9% vs.30.5%),P<0.01).The methylation rates of ZNF667-AS1 Cp G islands of various regions in RCC tissues were statistically analyzed which were grouped by the clinicopathological data(age,gender,tumor diameter,distant metastasis and TNM stage)of patients,and the difference was statistically significant(P>0.05).2.The expression of ZNF667-AS1 in RCC and corresponding normal tissues: the relative expression of ZNF667-AS1 mRNA in RCC tissues was lower than that in corresponding normal tissues significantly,and the difference was statistically significant(0.31(0.12,0.96)vs.1.00(1.00,1.00),P<0.001).The relative expression of ZNF667-AS1 mRNA in RCC tissues was statistically analyzed which was grouped by the clinicopathological data(age,gender,tumor diameter,distant metastasis and TNM stage)of patients,and the difference was statistically significant(P>0.05).3.The expression of ZNF667 in RCC and corresponding normal tissues: the relative expression of ZNF667 mRNA in RCC tissues was lower than that in corresponding normal tissues significantly,and the difference was statistically significant(0.42(0.11,1.09)vs.1.00(1.00,1.00),P<0.001).The relative expression of ZNF667 mRNA in RCC tissues was statistically analyzed which was grouped by the clinicopathological data(age,gender,tumor diameter,distant metastasis and TNM stage)of patients,and the difference was statistically significant(P>0.05).4.There correlation of expression of ZNF667-AS1 and ZNF667 mRNA in RCC tissues: there was a significant positive correlation between the relative expression of ZNF667 and ZNF667-AS1 mRNA in RCC tissues(r=0.775,P<0.001).Conclusion:1.The aberrant DNA hypermethylation of ZNF667-AS1 Cp G islands of the distal promoter,proximal promoter and exon 1 region in RCC may be related to the occurrence of RCC,but may not be related to age,gender,tumor diameter,distant metastasis and TNM stage of patients.2.The low expression of ZNF667-AS1 in RCC may be related to the occurrence of RCC,but may not be related to the age,gender,tumor diameter,distant metastasis and TNM stage of patients.3.The low expression of ZNF667 in RCC may be related to the occurrence of RCC,but may not be related to the age,gender,tumor diameter,distant metastasis and TNM stage of patients.4.There is a significant positive correlation between the expression of ZNF667-AS1 and ZNF667 mRNA in RCC,and there may be a certain synergistic effect in the occurrence and development of RCC. |
