Objective:In recent years,the morbidity and mortality of lung cancer are increasing year by year,which not only seriously endangers human health,but also causes a certain burden to the social economy.The treatment of lung cancer includes surgery,radiotherapy and chemotherapy,targeted drugs,immunotherapy,traditional Chinese medicine treatment and so on.However,because the early symptoms of most patients with lung cancer are not obvious,as soon as it is found that most patients have developed to the middle and late stage with metastasis,the chance of operation is less and the effect is poor.Radiotherapy,chemotherapy and targeted therapy are widely used at present,but the 5-year survival rate is not ideal.Hydrogen,as a hot medical gas in recent years,has the effects of anti-inflammation,anti-oxidation and anti-cancer,and has been found to have therapeutic effects on a variety of diseases.the purpose of this study is to explore the effect of hydrogen on apoptosis of human lung cancer A549 and NCI-H1975 cells,and to explore the role of STAT3/Bcl2 signaling pathway in it.Methods:1.Different concentrations of H2(20,40,60%)were used to interfere with lung cancer A549 and NCI-H1975 cells in logarithmic phase.Apoptosis was detected by flow cytometry(FCM),and Apoptosis-related proteins were detected by Western blot(WB).2.WB technique was used to detect the expression of STAT3,p-STAT3 and Bcl2 genes between the control group and hydrogen-treated lung cancer A549 and NCI-H1975 cells.3.STAT3 gene was overexpressed,and the transfection efficiency was detected by q PCR and WB.Apoptosis was detected by flow cytometry and apoptosis-related proteins were detected by WB.4.Immunohistochemistry was used to detect the expression of p-STAT3 and Bcl2 genes in metastatic tumor tissues of lung cancer A549 cells in nude mice,and the expression levels of hydrogen group,cisplatin group and control group were compared.5.Tunel method was used to detect apoptosis in control group,hydrogen group and cisplatin group.Results:1.Different concentrations of hydrogen can promote the apoptosis of lung cancer A549 and NCI-H1975 cells;2.Compared with the control group,the expression of STAT3 in A549 and NCI-H1975 cells treated with hydrogen had no significant difference(P>0.05),but the expression of p-STAT3 and Bcl2 decreased significantly(P<0.05);3.After overexpression of STAT3,hydrogen could inhibit the apoptosis of lung cancer A549 and NCI-H1975 cells(P<0.05);4.The expression of p-STAT3 and Bcl2 in the metastatic tumor tissue of lung cancer A549 cells in nude mice was detected by immunohistochemical method.Compared with the control group,the expression of hydrogen group and cisplatin group decreased significantly(P<0.05);5.Tunel method was used to detect the metastatic tumor tissue of lung cancer A549 cells in nude mice.Compared with the control group,the apoptosis in hydrogen group and cisplatin group was significantly higher than that in control group(P<0.05).Conclusions:1.Hydrogen can promote the apoptosis of lung cancer A549 and NCI-1975 cells.2.Hydrogen can promote apoptosis of lung cancer A549 and NCI-H1975 cells by inhibiting STAT3/Bcl2 pathway.Overexpression of STAT3 can reduce the effect of hydrogen on apoptosis of lung cancer cells.
