Objective:Hepatocellular carcinom(HCC)is a disease with high morbidity and mortality.Its recurrence and metastasis rate is high.In order to improve its therapeutic effect,it's particularly important to find new diagnosis and treatment targets.Long non coding RNA(lncRNA)is a kind of RNA which is longer than 200 nt and has no ability to encode protein.Previous studies have shown that lncRNA ADAMTS9-AS2 plays an important role in the occurrence and development of a variety of malignant tumors,however,its expression level and mechanism in HCC have not been reported.The purposes of this study are to investigate the expression of lncRNA ADAMTS9-AS2 in HCC and its effect on the development of HCC.Methods:1.The expression of lncRNA ADAMTS9-AS2 was detected by quantitative reverse transcription real-time fluorescent quantitative PCR(qRT-PCR)in 62 cases of HCC and normal tissues adjacent to the cancer.The correlation between the expression level of lncRNA ADAMTS9-AS2 and the clinicopathological characteristics of HCC was statistically analyzed.2.The expression level of lncRNA ADAMTS9-AS2 in three hepatoma cells(Huh7,SMMC-7721,QGY-7701)was detected by qRT-PCR,and was compared with that in 10 randomly selected normal tissues mixed up.3.LncRNA ADAMTS9-AS2 was overexpressed in SMMC-7721 cells by liposome transfection technology,and control group was set up.The transfection efficiency was verified by qRT-PCR technology.After transfection,the proliferation ability of SMMC-7721 cells was detected by MTS method,and the invasion ability of SMMC-7721 cells was detected by Transwell method.Results:1.The expression quantity of lncRNA ADAMTS9-AS2 is 0.027(0.009-0.719)in HCC tissues and is 0.053(0.021-0.172)in normal liver tissues,the difference is statistically significant(P < 0.05).2.The results showed that the expression level of lncRNA ADAMTS9-AS2 in HCC was not correlated with the age,gender,tumor size,tumor number and presence of microvascular tumor thrombus of HCC patients(P > 0.05).3.The expression quantities of lncRNA ADAMTS9-AS2 in the hepatoma cells Huh7,QGY-7701 and SMMC-7721 were 0.000158±0.000002,0.000257±0.000041,0.000373±0.000061,the expression quantity of the lncRNA ADAMTS9-AS2 in the 10 randomly selected normal liver tissues was 0.0115 ± 0.0013,the difference between cancer cell groups and normal group was statistically significant(P < 0.05).4.Compared with the control group,the proliferation activity and invasion ability of HCC cells were significantly reduced in the lncRNA ADAMTS9-AS2 overexpression group(P < 0.05).Conclusions:The down-regulation of lncRNA ADAMTS9-AS2 in HCC suggests that lncRNA ADAMTS9-AS2 may play a role as a tumor suppressor gene in the formation of HCC.When the lncRNA ADAMTS9-AS2 is over expressed,it can significantly inhibit the proliferation and invasion of HCC cells,suggesting that lncRNA ADAMTS9-AS2 is negatively related to the invasion and metastasis of HCC.
