ObjectiveThe number and activity of peritoneal macrophages in patients with endometriosis increased significantly,and the peritoneal macrophages were involved in the development of endometriosis.Hypoxia is an important factor in the development of endometriosis.This research will measure the oxygen partial pressure of the peritoneal fluid and explore the role of activation of NLRP3 inflammasome in peritoneal macrophages in the development of endometriosis.MethodsThis study collected peritoneal fluid from patients with and without endometriosis,measured the oxygen partial pressure of the peritoneal fluid,and isolated macrophages using ficoll to explore the difference of activation of NLRP3 inflammasome between the two groups.After the induction of hypoxia,the activation of NLRP3 inflammasome of macrophage was detected.Endometrial stomal cells were treated with peritoneal fluid and recombinant protein IL-1?to explore the influence of large amount of IL-1?secreted after the activation of NLRP3 inflammasome on the cell function of endometrial stromal cells.And endometriosis was surgically induced in NLRP3-/-and wild-type mice to identify the effect of NLRP3 on the development of endometriosis.Results1.The oxygen partial pressure of the peritoneal fluid in patients with endometriosis was significantly lower than that in the control group.The activation of NLRP3inflammasome in peritoneal macrophages of patients with endometriosis increased significantly;2.After the induction of hypoxia,the activation of NLRP3 inflammasome of macrophage was increased;3.Peritoneal fluid from endometriosis patients and IL-1?can promote proliferation and migration of endometrial stromal cells by activation of Akt-m TOR pathway;4.The ectopic lesions developed in NLRP3-/-mice were significantly smaller than those in wild-type mice.Intraperitoneal injection of macrophages derived from wild-type mice could promote the formation of endometriosis in NLRP3-/-mice.ConclusionHypoxia can induce the activation of NLRP3 inflammasome in macrophages.IL-1?secreted after the activation of NLRP3 inflammasome by macrophages can promote proliferation and migration of endometrial stromal cells by activation of Akt-m TOR pathway.The activation of NLRP3 inflammasome in peritoneal macrophages plays an important role in the development of endometriosis.
