Purpose: The survival rate of osteosarcoma(OS)patients has remained stagnant for decades.Given that OS has sophisticated multiomic heterogeneity and lacks pervasive targets,immune microenvironment,which is inclined to be less heterogenous,has gotten increasingly attention.For the present study,the database was used to find the prognostic genes which may tumor immune microenvironment?related for osteosarcoma.Methods: We used the data from Therapeutically Applicable Research to Generate Effective Treatments(TARGET).And via the ESTIMATE algorithm,immune scores were calculated to quantify the immune components in every sample.Further,based on sample immune scores,OS patients in the TARGET database were divided into two groups by the median and differentially expressed genes which are simultaneously genes associated prognosis were identified.And then functional enrichment analysis and protein?protein interaction networks analysis were performed to make further functional researches.And then,we validated these genes in an independent cohort from the Gene Expression Omnibus(GEO)database.Results: From TARGET,45 genes were recognized as hub tumor immune microenvironment?related prognostic genes which mainly get involved in inflammatory response,calcium ion homeostasis,immune response,chemokine signaling pathway and osteoclast differentiation.Combine with GEO database,finally,eight genes were recognized: PLCB4,SPI1,C3AR1,GPSM3,CD53,HCK,GNA15 and CD14.Conclusion: Eight genes abovementioned were preliminarily verified as critical immune microenvironment?related genes for osteosarcoma,which may become new potent targets.
