Objective: To investigate the effect of FETUB on the growth of prostate cancer and the possible mechanism.Methods: PCR,Western Blotting and immunohistochemistry were used to detect the difference expression of FETUB among the different cell lines(RWPE-1,PC-3,DU-145,LNCa P).PC-3 and DU-145 cell lines were selected as the research objects.we overexpressed FETUB in prostate cancer cells by lentivirus and we chose Western blotting to detect the expression level of FETUB after transfection.And cleaved-caspase3,Bcl-2,Bax were also detected by Western Blotting.In addition,flow cytometry detection was used to detected apoptosis,and using plate cloning to detect the effects of proliferation.Transwell and scratch test was used to detect the impact of invasion and metastasis.And using Western blotting to detect Vimentin and E-cadherin.Then PC-3 cell line was selected for nude mice tumor formation experiments to verify whether FETUB affects tumor proliferation in vivo.Results: FETUB was lowly expressed in prostate cancer cell lines and tissues.The index of prostate cancer overexpressing FETUB,cleaved-caspaese3,Bax,etc.increased,and Bcl-2 decreased.Flow cytometry showed an increase in this cell.Transwell results suggest that then Western blotting results showed that the expression levels of phosphorylated AKT and PI3 K in the protein of the overexpression group were reduced.The experiment of plate cloning showed that the growth rate of the overexpression group was slower.Nude tumor formation experiments showed that nude mice in the overexpression group had tumor formation.The speed decreases,and the tumor volume and weight are less than the minimum.
