ObjectiveTo explore the mechanism of qifuli Chinese on the signal of IL-4,IL-6,IL-10,TNF-?and m TOR/p-s6k1/p-4EBP1 in rats with ulcerative colitis?UC?.MethodSD male rats were randomly divided into two groups:normal control group?12 rats?and model group?60 rats?.The model rats were given 10mlˇkg-1 Rhubarb decoction for2 weeks.In the third week,TNBS ethanol enema of 100mg/kg-1 was used to establish UC model of spleen kidney yang deficiency.The model rats were randomly divided into model group(10mlˇkg-1ˇd),positive drug group(0.35gˇkg-1ˇd),high dose group(23.62ˇkg-1ˇd),medium dose group(11.81mgˇkg-1ˇd)and low dose group(5.91mgˇkg-1ˇd).The rats in each group were given corresponding drugs for 2 weeks,normal control group and model group with normal saline,positive drug group with sulfasalazine,Chinese medicine group with qifulizhong decoction.The normal control group was fed regularly,During the experiment,the macroscopical characteristics of rats in each group were recorded,and the body weight was monitored every week.The pathological changes of colonic mucosa were observed by HE staining,the levels of IL-4,IL-10,IL-6 and TNF-a in serum were detected by enzyme-linked immunosorbent assay,and the expression sites and levels of m TORC1,p-s6k1 and p-4ebp1 in colonic mucosa were detected by immunohistochemistry and Western blotting.The data were processed by spss20.0.Result1. Macroscopic characterization:the rats in the normal control group were active in spirit,which was generally acceptable;after the model was made by rhubarb+TNBS ethanol,the rats in the model were depressed in spirit,decreased in activity,serious depilation,decreased in food and water consumption,soft stool,increased in defecation times,and some rats had bloody mucus stool;after treatment,the general situation in each treatment group was improved.2. CMDI score:compared with the normal control group,the colonic mucosa of themodel group was significantly congested and edematous,with multiple ulcer lesions,and the CMDI score index was significantly increased?P<0.01?;compared with the model group,the CMDI score index of the positive drug group and the high-dose group of traditional Chinese medicine was significantly reduced?P<0.01 or P<0.05?,while the CMDI score index of the middle and low-dose groups of traditional Chinese medicine was decreased,but there was no significant difference Significance?P>0.05?.3. HE staining:the structure of columnar epithelium,lamina propria and muscularis of the colon mucosa in the normal control group was intact,the intestinal gland was regular,no denaturation and inflammatory exudation was found,and no hyperemia,edema and inflammatory infiltration was found in the submucosa;the epithelial cells of the colon mucosa in the model group were obviously necrotic and exfoliated,the glands in the lamina propria were significantly reduced,the goblet cells and crypts were reduced or disappeared,and the submucosa was edematous There were a lot of inflammatory cells infiltration in the bleeding and ulcer areas,and the pathological conditions of colon mucosa in each treatment group were improved to different degrees.4. Serum inflammatory factor level:the level of IL-4 in the serum of rats:compared with the model group,the level of IL-4 in the serum of rats in the normal control group,the positive drug group,the high and medium dose group of traditional Chinese medicine showed a significant increase trend?P<0.01 or P<0.05?.Although the level of IL-4 in the low dose group of traditional Chinese medicine increased,there was no significant difference?P>0.05?.The level of IL-10 in serum of rats:compared with the model group,the level of IL-10 in serum of rats in normal control group,positive drug group,high and medium dose group of traditional Chinese medicine showed a significant increase trend?P<0.01 or P<0.05?;although the level of IL-10 in serum of rats in low dose group of traditional Chinese medicine increased,there was no significant difference?P>0.05?.Level of IL-6 in serum of rats:compared with the model group,the level of IL-6 in serum of rats in normal control group,positive drug group,high,medium and low dose groups of traditional Chinese medicine showed a significant decrease trend?P<0.01?.TNF-a level in serum of rats:compared with the model group,the level of TNF-a in serum of rats in normal control group,positive drug group,high and medium dose groups of traditional Chinese medicine showed a significant decrease trend?P<0.01?.Although the level of TNF-a in serum of rats in low dose group of traditional Chinese medicine decreased,there was no significant difference?P>0.05?.5. Expression of m TORC1 protein in colonic mucosa of rats:compared with the model group,the expression of m TORC1 protein in colonic mucosa of rats in normal control group,positive drug group,high and medium dose group of traditional Chinese medicine showed a significant decreasing trend?P<0.01 or P<0.05?.The expression of m TORC1 in colonic mucosa of rats in low dose group of traditional Chinese medicine decreased,but there was no significant difference?P>0.05?.P-s6k1 protein expression in colonic mucosa of rats:compared with the model group,p-s6k1 protein expression in colonic mucosa of rats in normal control group,positive drug group and high-dose Chinese medicine group showed a significant decrease trend?P<0.01 or P<0.05?.Although the expression of p-s6k1 in colon mucosa of rats in the middle and low dose group of traditional Chinese medicine decreased,there was no significant difference?P>0.05?.P-4ebp1 protein expression in colon mucosa of rats:compared with the model group,the expression of p-4ebp1 in colon mucosa of rats in the normal control group and the positive drug group was significantly decreased?P<0.01?.Although the expression of p-4ebp1 in colon mucosa of rats in high,middle and low dose groups of traditional Chinese medicine decreased,there was no significant difference?P>0.05?.conclusionQifulizhong decoction can regulate the balance of pro-inflammatory factors and anti-inflammatory factors through m TOR signal pathway,effectively alleviate the inflammation of UC induced by TNBS,and improve and repair the colonic mucosa of UC rats.
