Diabetes mellitus is a chronic disease with elevated blood glucose as the main feature,and disorders of carbohydrate and fat metabolism caused by absolute or relative insufficient insulin secretion or insulin utilization disorder.In recent years,many animal experiments and clinical studies have shown that the tea and some of its biochemical ingredients that we generally consume have a blood sugar-lowering effect.However,in the current research on the hypoglycemic effect of many tea biochemical components,the research on the hypoglycemic effect of tea seed saponin has not yet been reported.There are few studies on the efficacy of saponin in lowering blood sugar.Therefore,this article will explore the hypoglycemic effect of the Theasaponin on the STZ-induced type 2 diabetes rat model.60 SPF adult male healthy Wistar rats weighing 200 ± 20 g were selected and randomly divided into 6 groups after a week of adaptive feeding: Blank control group(n=10)and Negative control group(diabetes model)Rats,n=10),Positive control group(metformin intervention group,n=10),Theasaponin low-dose intervention group(n=10),Theasaponin middle-dose intervention group(n=10),Theasaponin In the high-dose intervention group(n=10),a high-fat diet combined with intraperitoneal injection of streptozotocin(STZ)was used to establish a type ?diabetic rat model,and intervention feeding was performed for 40 days.Detect the fasting blood glucose,glucose tolerance,blood lipid,liver function and other indicators of rats,observe the tissue morphology,and study the effect of Theasaponin on blood glucose levels in diabetic rats;collect the intestinal contents in the cecum of rats in the test group,and pass 16 Sr DNA Sequencing analysis of intestinal flora changes in the cecum contents of diabetic rats under the intervention of Theasaponin;detection and exploration of glucose transporters(SGLT-1,GLUT-2)in the colonic intestine of diabetic rats under the action of Theasaponin by fluorescent quantitative PCR and changes in the expression of proglucagon gene(PLG),further explore the effect and mechanism of Theasaponin on the blood sugar level and intestinal homeostasis of STZ-induced type ? diabetic rats.The specific research results are as follows:(1)Theasaponin can effectively improve the symptoms of hyperglycemia and blood lipid metabolism disorder in type ? diabetic rats.After testing the blood glucose,blood lipid,liver function and other indicators of the test group,and systematically evaluating the effect of Theasaponin on blood glucose in diabetic rats,it was found that Theasaponin can effectively regulate the blood glucose levels of diabetic rats and improve the type 2 diabetes The levels of TC,TG,HDL-C,LDL-C and other related factors in mouse serum(P<0.01),reduce theactivity of AST and ALT in serum(P<0.01),repair the morphology of islet and liver tissues in diabetic rats.(2)Theasaponin can effectively regulate the structure and abundance of intestinal flora in diabetic rats.By collecting the contents of the cecum of rats in each experimental group and analyzing the bioinformatics by high-throughput sequencing technology,it was found that theasaponin has a certain regulation effect on the flora in the intestinal contents of diabetic rats.The intervention of theasaponin can increase the abundance of the intestinal contents of diabetic rats to a certain extent,increase the abundance of the dominant intestinal flora(such as Bacillus),and reduce the harmful flora(such as Bacteriodes).(3)Theasaponin can maintain the blood glucose level of rats by regulating the expression of glucose transporter and proglucagon gene.The expression of SGLT-1,GLUT-2,and PLGm RNA in the colons of rats in each experimental group was detected by fluorescent quantitative PCR.It was found that the expression of SGLT-1 and GLUT-2 genes in the colons of diabetic rats was significantly increased(P<0.01),And different doses of Theasaponin and metformin can inhibit expression upregulation due to diabetes mellitus to a certain extent(P<0.01).Compared with diabetic rats,the expression level of PLG in high,middle and low dose groups of Theasaponin was significantly increased(P<0.01),and the glucose reducing effect of PLG was improved.In summary,a certain dose of theasaponin(50-150 mg/kg)can maintain the blood glucose level in rats by regulating the glucose transporter and glucagon gene expression in diabetic rats,relieve the symptoms of hyperglycemia and dyslipidemia.The flora richness and homeostasis had significant intervention effects.
