Objective By detecting the expression level of PD-1 and T lymphocyte subsets in peripheral blood of patients with different pathological types and stages of non-small cell lung cancer,the correlation and clinical significance of the two were discussed.Detect the expression level of PD-1+on the surface of CD8+T cells in patients with non-small cell lung cancer,to explore the relationship between PD-1 and PD-1+CD8+T expression levels and clinical pathological indicators.Methods Retrospectively selected 50 patients with non-small cell lung cancer in our hospital from January 2019 to December 2019,all confirmed by cytology or histological pathology as NSCLC,this group was defined as a test group,taking 10ml of fasting venous blood in the morning.In addition,50 healthy medical examiners from our hospital during the same period were selected as the contrast group retrospectively,and 10ml of fasting venous blood in the morning was taken as the normal control of peripheral blood in the case group.Enzyme-linked immunosorbent assay?ELISA?method was used to detect the expression level of PD-1 in peripheral blood,and flow cytometry?FCM?was used to detect peripheral blood CD series cells?CD4+,CD8+,CD19+,CD16+56?NK??and PD-1+CD8+T expression ratio,immunohistochemical SP method to detect the expression level of PD-1 in tissues.Results1.The cell ratio of CD4+and NK in the case group were significantly lower than those in the healthy group,and the difference was statistically significant?P<0.05?;the expression levels of PD-1 and the cell ratio of CD8+in the peripheral blood were significantly higher than that in the healthy group,and the difference was statistically significant?P<0.05?;There was no statistically significant difference in the expression levels of CD4+,CD8+,NK,and peripheral blood PD-1 in patients with lung adenocarcinoma and lung squamous cell carcinoma?P>0.05?,and the difference inCD19+,between the two groups There was no statistical significance?P>0.05?.2.The ratio of CD4+cells in patients with lung adenocarcinoma and lung squamous cell carcinoma?b+?is higher than that in stage?+??t1=5.841,t2=4.032?,and the ratio of NK cells is higher than that in stage?+??t1=3.499,t2=3.250?,the ratio of CD8+cells is lower than that of stage III+IV?t1=4.604,t2=3.747?,and the expression level of PD-1 in peripheral blood is significantly lower than that of stage III+??t1=3.355,t2=3.169?.Statistical significance?P<0.05?;Therewas no statistically significant difference in CD19+,ratio?P>0.05?.3.Peripheral blood PD-1 and CD8+cell ratios of patients with lung adenocarcinoma and lung squamous cell carcinoma were significantly positively correlated?r1=0.642,r2=0.647?,and were significantly negatively correlated with NK cell ratio?r1=-0.567,r2=-0.630?,the differences were statistically significant?P<0.05?;there was no correlation with CD4+andCD19+,?P>0.05?.4.In terms of PD-1+CD8+T expression rate,the test group and the control group were?16.7±2.9?%,?7.9±1.0?%in order,the test group was higher than the control group,the difference was statistically significant??2=4.303,P<0.05?.5.The expression rates of PD-1 and PD-1+CD8+T are correlated with the pathological stage,lymph node metastasis,distant metastasis and tissue differentiation of patients with NSCLC?P<0.05?;but not correlated with gender,age and pathological type Sex?P>0.05?.Conclusion 1.Peripheral blood PD-1 expression in NSCLC patients is significantly increased,peripheral blood T lymphocyte subsets are correlated with PD-1 expression,and NSCLC pathological stage is also correlated,so peripheral blood T lymphocyte subsets combined with PD The detection of-1 expression level can better reflect the immune function of NSCLC patients,.2.PD-1 is highly expressed on the surface of CD8+T cells in NSCLC patients,and is related to pathological stage,lymph node metastasis,distant metastasis and tissue differentiation,suggesting that PD-1 molecules may interact with PD-L1 on tumor cells to Inhibiting CD8+T cell function and participating in NSCLC immune escape-related processes play a vital role in the occurrence and development of NSCLC.
