The Effect Of FANCI On Promoting Postoperative Recurrence And Metastasis Of Hepatocellular Carcinoma

ObjectiveThe morbidity and mortality of liver cancer are among the highest in China.With the continuous development of imaging,the rate of early diagnosis of liver cancer is getting higher and higher,and radical surgical resection is still the gold standard for treatment,but there are still a large number of patients with liver cancer found in the late stage,therefore,it is a difficult problem for us to find biomarkers for early diagnosis of liver cancer.So far,there is no research to explore the mechanism of FANCI in the occurrence and development of hepatocellular carcinoma.The purpose of this study is to understand whether the dysfunction of FANCI is involved in the pathogenesis of hepatocellular carcinoma,and how FANCI affects the proliferation,migration and invasion of hepatocellular carcinoma,which lays an experimental foundation for FANCI to become a diagnostic marker and a new target for treatment of hepatocellular carcinoma.MethodsWe used q RT-PCR method to determine the expression level of FANCI in HCC samples and cell lines,and combined with follow-up data and TCGA database to analyze the relationship between prognosis and prognosis.After we prepared si-FANCI transient Hep3 b cells and Huh7 cells,we used CCK-8 assay and colony formation test to determine the relationship between genes and cell proliferation;Transwell assay was used to evaluate cell migration and invasion;flow cytometry was used to detect tumor cell apoptosis;immunohistochemistry and Western blotting were used to identify the content of FANCI protein in tissue.Finally,the relationship between FANCI and tumor growth was verified by subcutaneous tumor foreign body implantation in nude mice.ResultsOur results showed that q RT-PCR detection showed that the expression of FANCI was significantly up-regulated in tissues and cell lines of HCC,which was closely related to the malignant characteristics and poor prognosis of HCC,and to the size of tumor.Subsequent experiments showed that knocking down FANCI could inhibit the growth of HCC cells in vivo and in vitro.Survival analysis showed that the high expression of FANCI in HCC patients led to worse prognosis.ConclusionIn this study,our experimental data showed that FANCI is an effective indicator for early diagnosis of HCC,and low FANCI can improve the prognosis of patients by inhibiting the proliferation of hepatocellular carcinoma cells.It can be seen that FANCI has potential value in clinical targeted therapy of lung cancer.
