Objective:Oral cancer is one of the common cancers,while tongue cancer is the most common oral cancer,most of which are squamous cell carcinoma,which is easy to occur at the lingual margin and has a high degree of malignancy.Due to the abundant blood circulation in the tongue body and frequent mechanical movements of the tongue,tongue squamous cell carcinoma(TSCC)has fast growth rate,strong infiltration and high metastasis rate.Cervical lymph node metastasis often occurs in the early stage,and even distant metastasis may occur,usually to the lung.Although the research on the mechanism of TSCC migration and invasion is gradually increasing and the diagnostic and therapeutic techniques for TSCC are also gradually improving,the incidence and mortality of TSCC are still increasing year by year,and better solutions are urgently needed clinically.In recent years,the theory of tumor stem cells has been increasingly promoted by people.Bone marrow mesenchymal stem cells(BMSCs)are considered as ideal bioengineered cells due to their wide source,self-renewal and proliferation,multidirectional differentiation potential,low immunogenicity,tumor tropism,etc.At present,a large number of studies show that BMSCs have different effects on the invasion of different tumor cells,either promoting or inhibiting,but there are few studies on the effect of BMSCs on TSCC.This study studies the effect of BMSCs on TSCC migration and invasion and the possible mechanism of action on tongue cancer through in vitro experiments.Methods:Tongue squamous cancer cell line cal-27 cells and hBM-MSCs cells were cultured in vitro,and the scratch test,cell migration test and cell invasion test were carried out respectively after the two cells were co-cultured.The effect of hBM-MSC on cal-27 migration and invasion was observed from the cell level.In order to clarify its mechanism of action,the relative mRNA expressions of cal-27 migration and invasion related genes E-cadherin,twist,slug,snail,MMP-2,MMP-9 after single culture and co-culture were detected by fluorescence quantitative PCR,and Western blot detection was focused on MMP-2,MMP-9,and the experimental data were statistically analyzed by SPSS 19.0 software.Results:Under the influence of hBM-MSCs,the number of Cal-27 cells migrating and invading obviously increased.The fluorescence quantitative PCR detection results showed that the E-cadherin gene expression of the co-cultured Cal-27 was down-regulated,the gene expression of twist,slug,snail,MMP-2,MMP-9 was up-regulated,and the Western blot analysis showed that the MMP-2,MMP-9 protein expression was up-regulated.It is speculated that hBM-MSCs may promote epithelial-mesenchymal transition and tumor growth by down-regulating E-cadherin and up-regulating twist,slug,snail,MMP-2,MMP-9 genes.Conclusions: Cal-27 and hBM-MSCs cells were successfully cultured and co-cultured with cal-27 and hBM-MSCs,providing the basis for subsequent experiments.Human bone marrow mesenchymal stem cells can promote the migration and invasion of tongue squamous cell carcinoma.
