Objective: To investigate the effect on apoptosis,membrane expression of calreticulin(CRT),and release of high mobility group box protein1(HMGB1)in oral squamous cell carcinoma cells by thermochemotherapy.Methods:1.The cells were divided into four groups,conventionally cultured cells were treated as control group,cells which treated with chemotherapy of pingyangmycin for 24 h were considered as chemotherapy group,and in hyperthermia group,cells were treated with hyperthermia for 60 min,and then conventionally cultured for another 24 h.Furthermore,in thermochemotherapy group,cells were treated with hyperthermia for 60 min after chemotherapy,and then conventionally cultured for another 24 h.2.CCK-8 assay was used to analysis the working concentration of pingyangmycin.3.Annexin V/PI staining and flow cytometry were used to analysis cell apoptosis after different treatments.3.Flow cytometry was used to analysis the expression of CRT after different treatments.4.ELISA was used to detect the secretion of HMGB1 after different treatments.Results:1.The working concentration were chosed for chemotherapy and thermo-chemotherapy.2.The effect of different treatments on the cell apoptosis of oral squamous cell carcinoma cells: after hyperthermia,chemotherapy and thermochemotherapy therapy,the cell apoptosis rate increased significantly.Futhermore,the thermochemotherapy group possessed significant elevation compared to chemotherapy group and hyperthermia group.3.The effect of different treatments on CRT membrane expression: after hyperthermia,chemotherapy and thermochemotherapy,the membrane expression of CRT increased significantly,and the difference was statistically significant.Futhermore,the thermochemotherapy group possessed significant elevation compared to chemotherapy group and hyperthermia group.4.The effect of different treatments on HMGB1 release: after hyperthermia,chemotherapy and combined therapy,the secretion of HMGB1 increased significantly,and the difference was statistically significant.Futhermore,the thermochemotherapy group possessed significant elevation compared to chemotherapy group and hyperthermia group.Conclusion:Hyperthermia,chemotherapy of pingyangmycin and thermochemotherapy could induce oral squamous cell carcinoma cells succumb to death,and at the same time,they can effectively induce membrane expression of CRT,and promote the secretion of HMGB1.Moreover,the combined application of hyperthermia and chemotherapy is significantly better than that of chemotherapy alone or hyperthermia alone in apoptosis induction,the expression of CRT and release of HMGB1.
