Objective: To investigate the influence and molecular mechanism of mi R-182 on the biological behavior of malignant pleural mesothelioma(MPM)cells.Methods: Analogue of human mi R-182(mi R-182 mimic)and NC-mimic were constructed.Human mesothelioma cells NCI-H2452 were divided into three groups: Control group(not transfected),NC-mimic group(transfected with NC-mimic),and mi R-182 mimic group(transfected with mi R-182 mimic).Then,the expression of mi R-182 in each group was determined by RT-PCR,cells viability was assessed by MTT,proliferation and apoptosis were detected by flow cytometry,and invasion ability was detected by Transwell.While Luciferase Assay and Western Blot were used to identify the downstream target molecules.Results: The expression of mi R-182 in the mi R-182 mimic group was significantly higher than that in the Control group and NC-mimic group(P< 0.001).The cell viability and proliferation of mi R-182 mimic group were significantly higher than those of control group and NC-mimic group.In addition,the number of cells passing through the transwell membrane in mi R-182 mimic group was significantly increased(P< 0.01).Luciferase reporter assays indicated mi R-182 mimic suppressed the activity of RECK 3?UTR,and there were lower RECK protein level,higher CCND1 and MMP-9 protein expression in the mi R-182 mimic group.Conclusion: Mi R-182 can increase the activity,proliferation and invasion ability of MPM cells under the downregulation of RECK expression,which revealing that mir-182 plays an important role in the occurrence and development of MPM.
