Objective: To investigate the potential mechanism of microrna-218(miR-218)involved in the invasion and metastasis of NSCLC by negatively regulating BCL9Methods: Human non-small cell lung cancer cell lines H460,A549,HBE,SK,H1299 were selected for cell culture.The expression of miR-218 in non-small cell lung cancer cell lines H460,A549,HBE,SK,H1299 was detected by qPCR.miR-218 analogue(miRNA mimic)and miRNA inhibitor(miRNA inhibitor)were transfected into six well plates by lipo3000 in non-small cell lung cancer lines H1299 and A549,respectively.After 48 hours,Western blot was used to detect the expression of BCL9 protein in the cell line.74 cases diagnosed as lung adenocarcinoma from 2014 to 2018 were randomly selected to detect the degree of BCL9 protein in tissues according to the requirements of immunohistochemistry kit.Results:1.The expression of miR-218 in non-small cell lung cancer cell lines A549 and H1299 was significantly different(P < 0.05).2.In A549 and H1299 cell lines,miR-218 inhibitor(miRNA inhibitor)and miRNA mimic(miRNA mimic)were transfected respectively.miR-218-inhibitor significantly increased the expression level of BCL9 protein in A549 cell line and miR-218-mimic significantly decreased the expression level of BCL9 protein in H1299 cell line.The difference was statistically significant(P < 0.05)3.Immunohistochemical results: the expression of BCL9 in lung cancer was significantly higher than that in peripheral normal lung tissues(P < 0.001),and the expression level of BCL9 was closely related to the main pathological subtype of the patient(P < 0.001),but not to lymph node metastasis(P = 0.173)Conclusion:1.miR-218 negatively regulated the expression of BCL9 in NSCLC.2.The high expression of BCL9 in NSCLC is related to clinicopathological factors...
