Objective: Lung cancer is one of the main causes of high morbidity and mortality of malignant tumors,and it is also a major public health problem that seriously threatens human health.Long non-coding RNAs(lncRNAs)are a new class of potential biomarkers and therapeutic targets for cancer.Previous Studies have shown that lncRNA PCAT1 plays an important role in the development of lung cancer.In this study,four SNPs in PCAT1(rs1026411 G>A,rs12543663 A>C,rs710886 T>C and rs16901904 T>C)were chosen to investigate the association between genetic variant in PCAT1 and susceptibility to lung cancer.Methods: This study was a hospital-based case-control study including 468 lung cancer cases and 561 cancer-free controls.Genotyping of SNPs was conducted by using Taqman? allelic discrimination methods.All statistical analyses were performed by IBM SPSS Statistics 22.Student's t test and ?2 test were used to compare the differences in the distribution of age,sex,and smoking exposure between the cases and the controls.Unconditional logistic regression was used to calculate odds ratios(ORs)and their 95% confidence interval(95%CI)to assess the association between PCAT1 polymorphisms and lung cancer risk.Multiplicative and additive models were used to explore the relationship between the genes-environmental interaction and lung cancer risk.Results: There were no any statistically significant associations between PCAT1 rs1026411,rs12543663,rs710886 and rs16901904 polymorphisms and lung cancer risk.However,polymorphisms in PCAT1 rs1026411 was observed to have significant associations with susceptibility to non-small cell lung cancer(AG vs.GG: OR=0.701,P=0.020 and AA+AG vs.GG: OR=0.711,P=0.017).PCAT1 rs710886 polymorphisms was observed to have significant associations with the risk of non-small cell lung cancer(CT vs.TT: OR=0.723,P=0.047 and CC+CT vs.TT: OR=0.729,P=0.038).Besides,PCAT1 rs1026411 polymorphism had a similar association with susceptibility to lung adenocarcinoma(AG vs.GG: OR=0.663,P=0.019 and AA+AG vs.GG: OR=0.685,P=0.020).Polymorphisms in PCAT1 rs710886 was observed to be associated with lung squamous cell carcinoma risk(CC+CT vs.TT: OR=0.638,P=0.040).PCAT1 rs16901904 Polymorphisms was associated with lung squamous cell carcinoma risk(CC vs.TT: OR=2.582,P=0.033 and CC vs.TT+CT: OR=2.381,P=0.048).In addition,there was no significant result in interactions between PCAT1 polymorphisms and smoking exposure on both additive and multiplicative models.Conclusions: PCAT1 rs1026411 was associated with susceptibility to non-small cell lung cancer and lung adenocarcinoma.PCAT1 rs710886 was associated with susceptibility to non-small cell lung cancer and lung squamous cell carcinoma.PCAT1 rs16901904 was associated with susceptibility to lung squamous cell carcinoma.The interaction between PCAT1 polymorphisms and smoking exposure had no significant association with lung cancer risk.
