Objective: Hepatocellular Carcinoma is a common malignancy with high mortality worldwide.At present,great breakthroughs have been made in the early diagnosis and treatment of liver cancer,such as surgical resection,local ablation,transcatheter arterial chemoembolization and liver transplantation,and great progress has been made in the treatment of liver cancer patients.However,metastasis and recurrence of tumor are very common in patients with liver cancer after operation,and the overall survival rate is still not optimistic.Currently,due to insufficient understanding of the pathologic molecular mechanism of HCC,effective treatment approaches are very limited.Therefore,it is crucial to find an effective biomarker that is important to guide the treatment of HCC.Long non-coding RNA(long non-coding RNA(LncRNA))is defined as a non-coding protein RNA molecule with a length greater than 200 nucleotides.Long non-coding RNA GACAT3(LncRNA GACAT3)has been reported to be involved in the development of breast cancer,colorectal cancer,non-small cell lung cancer and gastric cancer.However,whether it is associated with hepatocellular carcinoma has not been clarified.In this study,the expression of GACAT3 in liver cancer tissues and adjacent tissues was detected by qRT-PCR,and the relationship between GACAT3 and clinicopathological data was also investigated.Changes in proliferation,invasion and metastasis of HCC cells were detected by RNA interference,CCK8 assay and Transwell assay.Methods:62 cases of liver cancer and para-cancer tissues were collected,and the expression of GACAT3 in liver cancer tissues and para-cancer tissues was detected by qRT-PCR,and the relationship between GACAT3 and clinical pathological factors was analyzed.One normal liver cell line(LO2)and three liver cancer cell lines(HepG2,SMMC7721,Huh7)were selected and their expression was detected by qRT-PCR.The expression of GACAT3 in liver cancer was knocked out by RNA interference technique,and its transfection efficiency was detected by qRT-PCR.At the same time,the cck-8 proliferation kit was used to test the effect of cck-8 on the proliferation,metastasis and invasion ability of HCC cells by Transwell assay.Results: Through the qRT-PCR detection of 62 cases of liver cancer tissue and adjacent tissues and normal liver cell line and the expression of liver cancer cell lines,the cancer is cancer of the liver tissue adjacent tissues expression level increased significantly(P < 0.05),three kinds of liver cancer cell lines compared with normal liver cell line expressing content increased obviously(P < 0.05),and further analyzes its relationship with clinical pathology,found GACAT3 expression level is associated with tumor differentiation,lymph node metastasis and TNM stages(P < 0.05),and its has no obvious relation with age,sex and AFP level(P > 0.05).After transfection with GACAT3,the proliferation,invasion and metastasis of HCC cells were significantly reduced(P<0.05).Conclusion:GACAT3 is highly expressed in liver cancer tissues and cell lines,and its expression level is related to tumor differentiation,lymph node metastasis and TNM staging.The transfection interfered with the expression of GACAT3 and weakened the proliferation,invasion and metastasis ability of HCC cells.
