Effects Of Rhubarb On The Content Of MTL And GAS In Intestinal Tissue Of Rat Model Of Necrotizing Enterocolitis

Objective: The purpose of this project is to apply raw rhubarb to a necrotizing enterocolitis model of neonatal SD rats.By monitoring the general condition and weight changes of newborn rats,pathological scores of intestinal tissue,and motilin(MTL)and gastrin(GAS)content,to evaluate the therapeutic effect of raw rhubarb on necrotizing enterocolitis,and to provide experimental and theoretical basis for further application of this therapy in clinical practice.Methods: Seventy-two newborn SD rats aged 3 days were randomly divided into 6groups,12 in each group: blank control group(Group A),only receiving artificial feeding,no special treatment;the remaining 60 newborn rats divided into: model group(group B),cimetidine group(group C),high-dose raw rhubarb group(group D),medium-dose raw rhubarb group(group E),low-dose raw rhubarb group(group F).The model of necrotizing enterocolitis was established by artificial feeding,hypoxia for 90 s,reoxygenation for 10 min,cold stimulation at 4 ? and LPS injection.At the same time,the model group(group B)was given 0.9% NaCl injection treatment,the cimetidine group(group C)was given cimetidine suspension 0.01 mg / kg·times,the high-dose group(group D),the middle-dose group(group E)and the low-dose group(group F)were given raw rhubarb 1.32 mg / kg·times,0.66 mg / kg·times,0.33 mg /kg·times,all 0.1ml / times,3 times / day,3 consecutive days.The general condition and weight change of newborn rats were evaluated at the same time window of each day.The animals in each experimental group were fasted for 12 hours after the last feeding,and were sacrificed by cervical dislocation on the 4th day.The injured intestinal tissue was stained with HE and the pathological score of intestinal tissue was detected.The raw rhubarb was observed by examining the changes in the MTL and GAS levels for the treatment of necrotizing enterocolitis.Results: 1.General situation and weight change: B,C,D,E,F groups showed reduced activity,difficulty feeding,and gastric retention after modeling;irritability,convulsions,shortness of breath,and cyanosis after hypoxia.After reoxygenation,ittakes tens of seconds to 1 minute for groups D,E,and F to resume independent activities,and it takes about 3 minutes for groups B and C.After cold stimulation,the skin color recovery of groups D,E,and F is less than 1 minute,and for groups B and C,2-3 minutes.Comparison of weight gain between groups after treatment: A>E>D>F>C>B,(P> 0.05).2.HE staining:The intestinal tissue structure in group A was complete,the glands were arranged regularly,and there was no obvious congestion and edema in the mucosa and submucosa;the intestinal tissues in groups B and C were seriously damaged,intestinal villi degeneration,edema,gland arrangement disordered,and the submucosa and lamina propria were severely separated,With a large number of inflammatory cell infiltration;the intestinal tissue structure in groups D,E,and F is relatively neat,mild intestinal villi are congested and edema,the submucosa and lamina propria are slightly separated,and occasionally inflammatory cell infiltration;3.Intestinal histopathological score: Compared with group A,the NEC score of group B was significantly increased(P <0.05);compared with group B,the scores of groups D,E,and F were significantly decreased(P <0.05).There was no significant difference between group B and C(P>0.05).Compared with group C,the scores of groups D,E,and F were reduced(P <0.05);there was no significant difference in comparison scores between groups D,E,and F(P>0.05).4.Changes in MTL and GAS content in newborn rats: Compared with the model group,the MTL increased and GAS decreased in the raw rhubarb treatment group,and the differences were statistically significant(P <0.05);compared with the cimetidine group,The expression of MTL in the raw rhubarb treatment group was higher than that of the cimetidine group,and the GAS expression was lower than that of the cimetidine group,the differences were statistically significant(P<0.05);Comparison between different dose groups of raw rhubarb,the expressions of MTL and GAS were similar,and the differences were not statistically significant(P>0.05).Conclusion:Raw rhubarb can reduce the degree of intestinal injury in neonatal rat NEC model by promoting MTL expression and inhibiting GAS expression.
