Background Colorectal cancer,as a clinical multiple malignant tumor,is the fourth largest malignant tumor in the world and the third largest malignant tumor in China,which seriously threatens people's life and health.To explore the molecular regulatory mechanism of colorectal cancer is helpful to clarify its occurrence and development characteristics,and to provide scientific guidance for clinical diagnosis and treatment.In recent years,the academic community has paid special attention to the molecular regulation mechanism of patients with colorectal cancer,trying to explore new diagnostic methods and therapeutic targets of colorectal cancer by deepening relevant research.Micro RNA(micro RNA,micro RNA)is a kind of small single-stranded RNA with length of 18-25 nucleotides.It can participate in many important life activities through directional regulation of downstream target genes and play an important role in the occurrence and development of tumors.The relationship between micro RNA and tumors is a hot topic in the academic circles.Micro RNA-552 and micro RNA-592 are newly discovered micro RNAs.Researchers have found that micro RNAs-552 and micro RNAs-592 are significantly up-regulated in colorectal cancer tissues.It is speculated that micro RNAs-552 and micro RNAs-592 play an important role in the formation and development of colorectal cancer,but there are few related studies.Therefore,this study attempts to analyze the expression and clinical significance of micro RNA-552 and micro RNA-592 in colorectal tumors based on clinical experimental studies.Objective By comparing the expression of micro RNAs-552 and micro RNAs-592 in normal tissues of colorectal tumors and adjacent normal tissues,the expression of micro RNAs-552 and micro RNAs-592 in patients with different colorectal pathological characteristics,the expression of micro RNAs-552 and micro RNAs-592 in patients with different prognosis,the expression characteristics and significance of micro RNAs-552 and micro RNAs-592 in colorectal tumors were analyzed in order to provide a basis for clinical diagnosis,evaluation and treatment of colorectal tumors.For reference.Methods 79 patients with colorectal cancer who underwent surgical excision and reserved pathological specimens from January 2014 to December 2015 in Cancer center of Guangzhou medical university Hospital were studied.All patients were followed up for 3 years(from 3 years to death).Real-time fluorescence quantitative(RT-PCR)was used to detect the levels of micro RNAs-552 and micro RNAs-592 in tumor tissues and adjacent normal tissues of all patients.The expression of micro RNAs-552 and micro RNAs-592 in tumors and normal tissues were compared.On this basis,the expression of micro RNAs-552 and micro RNAs-592 in pathological tissues of patients with different pathological characteristics and prognosis were compared.The relationship between the expression levels of micro RNAs-552 and micro RNAs-592 and pathological tissues and prognosis of colorectal tumors was analyzed by logistic multivariate regression analysis.Results(1)The expression of micro RNA-552 in tumor tissue was significantly higher than that in normal tissue(P < 0.05).The expression level of micro RNA-552 in patients with different TNM stages,differentiation degree and lymph node metastasis was significantly different(P < 0.05).The higher TNM stage,the higher expression level of micro RNA-552;the lower differentiation degree,the higher expression level of micro RNA-552;the higher expression level of micro RNA-552 in patients with lymph node metastasis than in patients without lymph node metastasis;the lower expression level of micro RNA-552 in patients with different gender,age and tumor length,the lower expression level of micro RNA-552.There was no significant difference(P > 0.05).The expression level of mi R-552 in patients with recurrence or death within 3 years was significantly higher than that in patients with disease-free survival within 3 years(P < 0.05).Logistic multivariate regression analysis showed that the expression of micro RNA-552 was closely related to TNM stage,differentiation degree,lymph node metastasis and prognosis,which was positively correlated with TNM stage and lymph node metastasis,and negatively correlated with differentiation degree and prognosis.(2)The expression of micro RNA-592 in tumor tissue was significantly higher than that in normal tissue(P < 0.05).The expression of micro RNA-592 in patients with different TNM stages,differentiation degree and lymph node metastasis was significantly different(P < 0.05).The higher TNM stage,the higher expression level of micro RNA-592;the lower differentiation degree,the higher expression level of micro RNA-592;and the higher expression level of micro RNA-592 in patients with lymph node metastasis than in patients without lymph node metastasis.There was no significant difference in the expression of micro RNA-592 between patients with different gender,age and tumor length(P > 0.05).The expression level of micro RNA-592 in patients with recurrence or death within 3 expression levels of micro RNA-552 and micro RNA-592 in colorectal cancer tissues were closely related to different TNM stages,differentiation,lymph node metastasis and prognosis.It is suggested that the expression of micro RNA-552 and micro RNA-592 can be used to detect and diagnose colorectal tumors and evaluate their occurrence,development,metastasis and prognosis.
