Objective:At different time points,the level of CGRP in the vascular wall of the rat model of subarachnoid hemorrhage was detected by immunohistochemistry and enzyme-link-ed immunosorbent assay.The morphological changes of the basilar artery were observed by HE staining.The apoptosis of neurons were observed under electron microscopy,and intervene with butylphthalide injection to preliminary explore the effect of butylphthalide injection on CGRP and neuronal apoptosis after subarachnoid hemorrhage.Method:Forty-five SD(Sprague dawley)male rats weighing 220-270 g were selected.Subsequently,the rats were randomly divided into three groups using the random number method:SAH(subarachnoid hemorrhage)group(N=15),SAH+NBP(subarachnoid hemorrhage+butylphthalide injection)group(N=15),NS(normal saline)group(N=15).In the SAH group,a subarachnoid hemorrhage model was established using the blood injection method of the optic chiasm.The SAH+NB P group was injected butylphthalide injection by intraperitoneal at30 minutes after the successful modeling,the total amount was 5m g/kg/d,divided into two for injection,the NS group was injected an equal amount of saline into the anterior optic chiasm.Each groupis randomly divided into 3 subgroups:1 day group(N=5),3 day group(N=5),5 day group(N=5),1 day,3 days,5 days subgroup in SAH+NBP group received continuous intraperitoneal injection of NBP for 1 day,3 days,and 5 days.Each group was underwent neurological function scoring and calculated the weight loss ratio at the first day after the model was established.The value of serum CGRP were de-termined by ELISA at predetermined time points.The brain was taken from the head and the brain tissue was taken for HE staining to observe the morphologyofthe basilar artery and the expression of CGRP in the basilar artery.One rat hippocampus was randomly selected from SAH and SAH+NBP 1 day subgroups,and neuron apoptosis was observed under electron microscope.Results:1.Nerve function score,weight loss ratio:The postoperative neurological function score of SAH group was significantly higher than that of SAH+NBP(P<0.05).NS?SAH?SAH+NBP groups have a significant difference in weight loss ratio(P<0.05),SAH group weight loss ratio is higher than SAH+NBP group(P<0.05).2.The values of CGRP in serum:The CGRP of SAH,SAH+NBP and NS groups had significant differences(P<0.05).The NS group wass ignificantly higher than the SAH group(P<0.05).The SAH+NBP group was higher than the SAH group(P<0.05),and the serum CGRP of t-he two groups reached the lowest value on the third day.3.HE staining:NS group showed no vasospasm of the basilar artery,intact endothelial cells,smooth tube wall;SAH group showed reduced inner diameter of thebasilar artery,swollen endothelial cells,thickened tube wall;the SAH+NBP group had a reduction in the diameter of the basilar artery,endothelial swelling,thickened tube wall,compared with the SAH group.4.Immunohistochemical results:The difference in the expression of CGRP in the basilar artery wall of the NS?SAH?SAH+NBP group was statistically significant(P<0.05).Compared with the SAH,S AH+NBP group,the NS group had significantly increased CGRP expression in the basilar artery wall(P<0.05),the expression of CGRP in the basilar artery wall in the SAH+NBP group was significantly higher than that in the SAH group(P<0.05).5.Neuron apoptosis:SAH group can be observed under the electron microscope:it shows the typical neuronal apoptosis morphological characteristics,while SAH+NBP group does not seen the typical neuron apoptosis structure.Conclusion:1.Butylphthalide injection has an important neuroprotective effect after subarachnoid hemorrhage in rats.2.Butylphthalide injection may improve the early prognosis aftersubarachnoid hemorrhage by increasing CGRP expression and reducing neuron apoptosis.
