Objective:(1)To describe the carrier status of HHV-6 among the elderly population in a district of Shenzhen who underwent physical examination;(2)To analyze the differences between elderly population with normal cognitive function and early cognitive dysfunction regarding their HHV-6 positive carrier rate;(3)To explore the potential influence of HHV-6 carrying rate and carrying amount on different cognitive function fields,including memory,attention and computational power.Methods:(1)The elderly people who participated in a physical examination in Shenzhen from March to August 2018,were selected in a cluster sampling.The questionnaire survey and blood collection were carried out,and the database was established by using Epi Data software;(2)According to MMSE diagnostic criteria,the elderly were divided into case group and control group,before being matched at a ratio of 1:1 into the study according to gender and age(± 3 years old);(3)The HHV-6 gene was detected by real-time fluorescence quantitative PCR-TaqMan probe method.The relationship between HHV-6 carrying and cognitive dysfunction was analyzed by multivariate linear regression,multivariate conditional logistic regression,and restrictive cubic spline.Results:(1)A total of 580 elderly people were included and there was no significant difference in age,gender,and education between the two groups(P>0.05).(2)There was no significant difference in the distribution of the HHV-6 gene between the two groups(OR=1.419,95%CI:0.799~2.521).After adjusting gender,age,education level,lunch break in the past half-year,sleep quality,self-rated depression symptoms,the ability of daily life,Hcy and WBC,mono,EO,BaSO and HbA1 c converted by lg,the results showed that HHV-6 was inversely correlated with the arithmetical abilities and attention of the elderly in physical examination(B=-0.385,P<0.05).(3)After adjusting gender,age,education level,sleep quality,Hcy,and HbA1 c converted by lg,the results of multiple linear regression showed that there was a statistically negative correlation between lg-converted HHV-6 copy number and multiple abilities regarding orientation(B=-0.974,P=0.013),arithmetic and attention(B=-1.840,P<0.001),language(B=-0.647,P<0.001),understanding and execution(B=-1.620,P=0.004).(4)There is a linear dose-response relationship between HHV-6 carrying capacity and orientation force(B=-0.893,95%CI:-1.881~0.0947),and a nonlinear dose-response relationship between HHV-6 carrying capacity and arithmetic and attention(B=-3.142,95%CI:-4.236~-2.048),language(B=-0.685,95%CI:-1.155~-0.216),understanding and capabilities(B=-1.580,95%CI:-2.990~-0.170).Conclusion:(1)The prevalence of HHV-6 in the elderly is generally low in Shenzhen;(2)In the subject population,there was a negative correlation between carrying rate of HHV-6 and computation attention,and there was a negative correlation between carrying amount of HHV-6 and orientation,computation attention,language ability,and understanding execution.(3)In the subject population,there was a linear dose-response relationship between HHV-6 carrying capacity and orientation abilities,and a nonlinear dose-response relationship between HHV-6 carrying capacity and arithmetic,attention,language ability,understanding,and execution capabilities.
