| Objective:Peritoneal fibrosis is one of the long-term complications of patients with peritoneal dialysis,which can lead to the failure of peritoneal dialysis.Recent studies have shown that spleen tyrosine kinase is associated with the occurrence of various fibrosis diseases,such as renal fibrosis and liver fibrosis.However,there is no information on the effect of spleen tyrosine kinase on peritoneal fibrosis.In this study,by observing the expression of splenic tyrosine kinase in peritoneal tissue of rat model of peritoneal fibrosis,it aims to explore the role of spleen tyrosine kinase in peritoneal fibrosis,and look for new therapeutic targets for peritoneal fibrosis.Methods:Twelve healthy adult SD male rats were selected and randomly divided into two groups,6 in the normal group without special treatment;while 6 rats in the chlorhexidine group,were treated intraperitoneally with 0.1%chlorhexidine of 10ml/kg for 3 weeks to establish a model of peritoneal fibrosis.After the modeling,the rats were euthanized at the same time.The peritoneal tissue and blood samples were retained.Then,the peritoneal tissue was stained with HE and Masson to observe the histopathological changes of the peritoneum.And immunohistochemical and quantitative reverse transcription polymerase chain reaction(qRT-PCR)were used to detect the expression of SyK.The levels of interleukin-1β(IL-1β),tumor necrosis factor-α(TNF-α),interleukin-6(IL-6)and monocyte chemoattractant protein-1(MCP-1)in blood were measured by enzyme-linked immunosorbent assay(ELISA),and the correlation was compared with the expression of SyK mRNA in the model group.Results:1.The peritoneal fibrosis model was successfully established by intraperitoneal administration of chlorhexidine.2.The results of immunohistochemistry and q RT-PCR showed that compared with the normal group,the peritoneal tissues of the model group highly expressed spleen tyrosine kinase(immunohistochemistry:2.80±0.42 VS 5.50±0.97,P<0.05;q RT-PCR:1.21±0.73 VS 4.97±2.05,P<0.01).3.The results of serum inflammatory cytokines IL-1β,TNF-α,IL-6 and MCP-1 measured by ELISA indicated that compared with the normal group,the levels of four inflammatory cytokines were increased in the model group(1.71±0.53ng/mL VS 6.76±2.72ng/mL,P<0.01;1.23±0.60ng/mL VS 4.21±2.13ng/mL,P<0.01;1.15±0.71ng/mL VS10.69±3.15ng/mL,P<0.01;1.59±0.67ng/mL VS 6.98±3.65ng/mL,P<0.01).4.Pearson correlation analysis of SyK mRNA level and inflammatory cytokines in model group showed that the expression of spleen tyrosine kinase was positively correlated with the levels of four inflammatory cytokines(R2=0.7462,P=0.0265;R2=0.7796,P=0.0198;R2=0.7807,P=0.0195;R2=0.7635,P=0.0229).Conclusion:The expression of SyK in peritoneal tissue of chlorhexidine-induced peritoneal fibrosis rat model was increased,which was positively correlated with the level of inflammatory factors.Spleen tyrosine kinase may be involved in the process of peritoneal fibrosis,which may provide new therapeutic targets for the prevention and treatment of peritoneal fibrosis. |
