Objective:To study the difference in the expression level of Retinoic Acid Induced 14(RAI14)gene in gastric cancer tissues and normal tissues adjacent to the cancer tissues The relationship between the different expression levels in gastric cancer tissues and the overall prognosis of gastric cancer patients.Methods:From January 2019 to March 2019,42 specimens of cancerous tissues and normal adjacent tissues(distance from the edge of the mass>5cm)of gastric cancer patients were collected and analyzed by Quantitative Real-time PCR qRT-PCR method to detect RAI14.Relative expression of m RNA.Another sample of 122 patients with gastric cancer tissues and adjacent normal tissues(distance from the edge of the mass>5cm)of122 patients who underwent radical gastric cancer D2 surgery at Anhui Provincial Hospital from March 2011 to May 2013 were selected by immunohistochemistry(Immunohistochemistry)to detect the expression of RAI14 protein.The chi-square test was also used to analyze the different expression levels of RAI14 gene in gastric cancer tissues and the clinicopathological characteristics of patients(for example:the age of the patient,whether the patient is male or female,whether the tumor pathology is a low-differentiation type or a medium/high-differentiation type,tumors Whether the diameter is greater than 5 cm,the tumor tissue infiltrates the gastric tissue(Tx),and the number of peripheral lymph node metastases(p N).The survival curve of the relationship between the expression of RAI14 and the overall survival(OS)of gastric cancer patients was drawn.The relationship between the expression of RAI14 in gastric cancer tissues and the prognosis of patients was analyzed by a single factor;the independent factors affecting the prognosis of gastric cancer patients were analyzed by multiple factors.Results:The qRT-PCR results showed that the expression of RAI14 m RNA in gastric cancer tissues was significantly higher than the corresponding expression level in normal adjacent tissues(t=4.976,P<0.001).The results of immunohistochemical detection showed that the expression level of RAI14 protein in gastric cancer tissues was higher than that in normal tissues adjacent to the cancer(72.2%vs 30.4%,?~2=42.665,P<0.001)..The survival time of RAI14 negative patients was significantly better than that of positive patients(?~2=10.939,P<0.001).The pathological features of gastric cancer tumors are poorly differentiated,whether the diameter of the tumor is larger than 5 cm,which layer of gastric tissue is infiltrated into the gastric tissue,and the number of peripheral lymph node metastases are closely related to the high expression of RAI14 protein.In terms of angle,the difference is significant(P<0.05).According to univariate analysis,the number of peripheral lymph node metastases,tumor diameter,abnormal expression of RAI14 gene,depth of tumor invasion,and degree of tumor pathological differentiation were correlated with the overall survival time of patients with gastric cancer(P<0.05).The age of gastric cancer patients,whether male or female,was not significantly related to the overall survival of patients.We further obtained through Cox multivariate judgment results:the independent factors affecting the overall prognosis of gastric cancer patients were abnormal expression of RAI14 gene,the number of metastases in peripheral lymph nodes,and which layer of gastric tissue infiltrated into gastric tissue(P<0.05).Conclusion:The occurrence and development of gastric cancer is related to the high expression of RAI14 gene in gastric cancer tissues.In terms of evaluation and monitoring of the prognosis of gastric cancer patients,RAI14 is expected to become a new biological marker for gastric cancer.
