ObjectiveLymphocytes are highly sensitive to radiation.Innate lymphoid cells?ILCs?are a type of immune cells with lymphocyte morphology but lacking specific antigen recognition receptors.ILCs are considered to he "minor cells" of helper T cells in the innate immune system.It is divided into cytotoxic ILCs?NK cells?and auxiliary ILCs?ILC1,ILC2,ILC3?according to the Strength of the tilling ability.Helper ILCs are present in lymphoid organs,are enriched in the mucosa and barrier surfaces,and may also be present in circulating peripheral blood.As innate immune cells,they play a very important role in the early stages of the immune response.In recent years,ILCs have been recognized as important mediators of immune homeostasis and pathology,and have played an important role in human tissue remodeling and repair.Coronary heart disease has become the disease with the highest mortality rate among chronic non-infectious diseases in the world.Atherosclerosis?AS?is the main cause of coronary heart disease.Lipid metabolism disorders are the basis of AS.Inflammatory reactions exist in arteriosclerosis.At various stages,lymphocytes play an important immune response.With the continuous development of medical intervention technology,percutaneous coronary intervention?PCI?under the guidance of X-iay fhioioscopy has become one of the main methods of coronary heart disease vascular Teconstruction with characteristics of less trauma and good curability.In various imaging methods,because PCI requires longer time,the radiation dose to patients also increases,and radiation exposure during interventional procedures has attracted more and more attention.Therefore,it is urgent to find out whether radiation will affect the therapeutic effect of coronary heart disease patients during interventional treatment,in order to provide better clinical guidance for coronary heart disease patients,and to provide new ideas for studying the immune regulation mechanism of coronary heart disease.However,it is unclear whether there is a direct relationship between ILCs and coronary heart disease.Whether X-ray radiation exposure during interventions has an impact on coronary heart disease through ILCs requires further study.The purpose of this study was to investigate the changes in the rate of ILCs in patients with coronary heart disease under X-iay radiation.The changes may inspire better clinical guidance during interventional treatment,that also may suggested mat patients should be given better the importance of radiation protection.Method?1?In this study,the peripheral blood of L4 healthy people and 20 patients with coronary heart disease undergoing interventional surgery were collected before and after interventional surgery.Mouse anti-human monoclonal fluorescent antibodies CD45,CD127,CD117,CD294,Lineage?CD3/14/19/20/56/11b/11c?line flow cytometry?FCM?was used to detect ILCs expression,and healthy people and patients with coronary heart disease?Preoperative?Triglyceride?TG?,low density lipoprotein cholesterol?LDL-C?,and total cholesterol?TC?levels in serum.?2?Mice were irradiated with a similar dose of X-rays during simulated inlerventional surgery.Flow cytometry was used to detect the expression of peripheral blood ILCs before and after irradiation,and serum ELISA was used to detect IL-12 levels.?3?APQE-/-mice were fed a high-fat diet for 10 weeks,and the ratio of ILCs in peripheral blood was detected by flow cytometry;TG,TC,and LDL-C levels in serum of APOE-/-mice before and after X-ray irradiation were biochemically measured.Reults:?1?Compared with the coronary heart disease?preoperative?group,the ILC1 expression rate of the coronary heart disease?postoperative?group increased and the ILC3 expression rate decreased;the ILC1 expression rate of the coronary heart disease?preoperative?group?89.42 ± 3.21%?,ILC3 The expression rate?6.54 ± 2.8%?and ILC1 expression rate?93.32 ± 3.39%?and ILC3 expression rate?4.42 ± 2.73%?in the coronary heart disease?postoperative?group were higher than those in the coronary heart disease?postoperative?group.The expression rate of ILC3 decreased by 4.4%,and the expression rate of ILC3 decreased by 32.4%.The difference was statistically significant?P<0.05?.?2?Compared with the control group,the peripheral blood ILC 1 rate of the mice in me irradiation group increased:the control group ILC1 rate?22.8 ± 5.5%?and the irradiation group ILC 1 rate?30.5 ± 4.02%?compared with the irradiation group;The rate of mouse ILC1 increased by 33.8%?P<0.05?,and the difference was statistically significant.At the same time,the concentration of IL 12 in serum of mice in the irradiation group was higher than that in the control group,?P<0.05?,and the change trend was consistent with the rate of ILC1.?3?Compared with the healthy control group-the ILC1 rate increased and the 1LC3 rate decreased in the coronary heart disease?preoperative?group:healthy control group ILC1?78.9 ± 4 37%?,ILC3?16.64 ± 6 6%?,and coronary heart disease?operation Before?Compared with ILC1?8942 ± 321%?and ILC3?654 ± 2.8%?,the ILC1 rate in the coronary heart disease?preoperative?group increased by L33%,and the ILC3 rate decreased by 60.7%?P<0 01?was statistically different.?4?The levels of LDL-C,TC,and TG?4.41 ± 0.12mmol/L,6 86 ± 0.75mmol/L,2.12 ±0.51mmol/L?in the coronary heart disease?preoperative?group were higher than those in the healthy group.Level?respectively 3.78 ± 0.13mmol/L,4.65 ± 0.96mmol/L,1.12± 0.51mmol/L?,P<0.05,the difference was statistically significant;by the healthy control group and coronary heart disease?preoperative?group Spearman correlation analysis was performed on ILCs and serum lipid indicators such as low-density lipoprotein cholesterol?LDL-C?,total cholesterol?TC?,and triglycerides?TG?.The rate of ILC1 was positively correlated with LDL-C.TC,and TG Sex?R2 was 0.54,0.79,0.56,P<0.001?.?5?Compared with the WT group,the rate of ILC1 in the APOE-/-group increased:the rate of ILC1 in the WT group?22.8 ± 5.5%?and the rate of ILC1 in the APOE-/-group?37.2 ± 2.78%?compared with APOE-/-The rate of ILC1 in the mice increased hy 63.2%?P<0.001?.and the difference was statistically significant.?6?X-rays did not significantly change serum lipid parameters TC,TG,and LDL-C in APOE-/-mice.Conclusion:In patients with coronary heart disease whose ILCs have been imbalanced,X-rays during intervention again increase the ILC1 expression rate,which may increase the risk of coronary heart disease development.
