BackgroundBreast cancer refers to malignant tumors that occur in the epithelial tissues of the breast,and the incidence of breast cancer worldwide has been increasing in recent decades.Breast cancer is the most common malignant tumor among Chinese women.The incidence of breast cancer has increased in recent years,while the survival rate of breast cancer patients has also been extended at the same time.Post-transcriptional gene regulation(PTGR)is essential for maintaining cellular metabolism and coordinating various RNAs through regulating their maturation,transport,stability,and degradation.Each of these events is regulated by the formation of different ribonucleoprotein(RNP)complexes,which have an RNA binding protein(RBP)at their core.So far,comprehensive literature reports that there are about 1,900 types of human RNA-binding proteins.PCBP2 plays a core role in m RNA stabilization and translation enhancement,thereby promoting the development and progress of human cancer.The protein of PCBP2 with multiple functions and functions as a stable complex with PCBP-1 and hn RNPK.It is one of the major cellular poly(r C)binding proteins.PCBP2 contains three K homology(KH)domains that may be involved in RNA binding,and plays a role in the formation of sequence-specific ?-globin m RNP complexes related to the stability of ?-globin m RNA.However,the current role of PCBP2 in the occurrence and development of breast cancer are still unclear.AimTo explore the role of PCBP2 in human breast cancer development and progression.Methods1)The expression of PCBP2 protein in breast cancer tissues and adjacent tissues were detected by Immunohistochemistry(IHC);the expression of PCBP2 in normal breast epithelial cell lines and breast cancer cell lines were detected by Western Blot;2)Explore the correlation between PCBP2 protein expression and clinicopathological parameters by analyzing clinical data;analyze the correlation between PCBP2 expression and disease progressionof breast cancer patients by using Kaplan-Meier survival curves;3)Two breast cancer cell lines were selected for stable transformation and cell experiments were performed.The MDA-MB-231 cell line with high PCBP2 expression was established for knockdown,and the SUM159 cell line with low PCBP2 expression was established for stable expression.After the establishment of the cell line,observe the changes in cell viability,proliferation,and migration and invasion capabilities through cell viability experiments,clone formation experiments,migration and invasion experiments.Result1)IHC results showed that the protein expression level of PCBP2 is overexpression in breast cancer tissues compared with adjacent tissues(P< 0.05);Western Blot results showed that protein of PCBP2 is overexpression in breast cancer cell lines compared to normal breast epithelial cell line;2)The overexpression of PCBP2 protein was significantly positively correlated with tumor size,lymph node metastasis and advanced clinical stage;The expression of PCBP2 may be significantly associated with disease progression and poor prognosis in breast cancer patients(P< 0.05);3)Cell viability experiments,clone formation experiments,migration and invasion experiments showed that the knockdown of PCBP2 reduced the proliferation,migration,and invasion of breast cancer cells MDA-MB-231(P< 0.05);while the overexpression of PCBP2,promoted proliferation,migration and invasion of breast cancer cells SUM159(P< 0.05).ConclusionPCBP2 plays an important role in breast cancer development and progression,and could be a potential biomarker for breast cancer treatment and diagnosis.
