Glycolipid metabolism disorders become an important factor in inducing cardiovascular disease,and its incidence is increasing day by day.At present,a variety of chemical drugs are often used in clinical treatment.Patients with long-term medication will have liver and kidney damage and easy to rebound after withdrawal and other side effects.And there is no means to prevent the disorder of glucose and lipid metabolism.Finding safe and effective drugs for the prevention and treatment of glucose and lipid metabolic disorders is of great significance to reduce the risk of cardiovascular disease.Hawthorn fruit has the function of eliminating food and invigorating stomach,resolving turbidity and reducing lipid,and Hawthorn leaves have cardiovascular protective effect.In this paper,the rat model of glucose and lipid disorders metabolism and the rat model of glycolipidemia were established to investigate the synergistic effect of Hawthorn fruit,leaves and their combination on the prevention and treatment of glycolipid metabolism disorders.The contents of the study are as follows:1.The preventive effect of hawthorn fruit organic acids,hawthorn leaves flavonoids and their combined use on glucose and lipid metabolism disorders were evaluated.Rats model of glucose and lipid metabolism disorders were established by diet induction,at the same time,hawthorn fruit organic acids,hawthorn leaves flavonoids and there combined use were given for 4 weeks.The results showed that the rats induced by high-fat and high-sugar diet had the phenomenon of weight gain,dyslipidemia,high blood glucose,abnormal liver and kidney function,which indicated that the rats model were established successfully.Compared with the model group,it was found that all of the drugs can antagonize the weight gain,high blood lipid,high blood glucose and abnormal liver and kidney dysfunction induced by diet,and the combination of hawthorn fruit and leaves was significantly better than hawthorn fruit and leaves alone in reducing blood glucose,TG,ALT,AST and UA.The results showed that hawthorn fruit organic acids and hawthorn leaves flavonoids have obvious effect on preventing the disorders of glucose and lipid metabolism,and the combination of fruit leaves was better than that of hawthorn fruit and leaves alone.2.To study the therapy effect of hawthorn fruit organic acids,hawthorn leaves flavonoids and their combined use on glucose and lipid metabolism.Rats with glucose and lipid metabolism disorders induced by high-fat and high-sugar diet combined withstreptozotocin were given different extracts of hawthorn for 7 weeks.The results showed that the rats induced by diet intervention combined with STZ appeared the phenomenon of hyperglycemia,impaired oral glucose tolerance,dyslipidemia,fat accumulation,morphological disorder of adipocytes,abnormal liver and kidney function,which indicated that the rats model were established successfully.It was found that different extracts of hawthorn reduced fasting blood glucose,enhanced glucose tolerance and regulated abnormal glucose metabolism.It also reduced liver index,improved blood lipid levels,protected fat tissue morphology,and regulated lipid metabolism disorders.It also protect liver and kidney functions.Moreover,hawthorn leaves used alone are better than hawthorn fruit in reducing ALP,CREA,UA and promoting insulin secretion,hawthorn fruit used alone are better than hawthorn leaves in reducing TG,body fat weight and TBIL.The combination group of hawthorn fruit leaves are significantly superior to the single administration group in reducing TG,AST,ALP,UA and improving the morphology of adipose tissue.It is found that hawthorn leaves flavonoids has better effect on regulating glucose metabolism,while hawthorn fruit organic acids improved fat accumulation and lipid metabolism disorder are better than hawthorn leaves flavonoids,and the combination of hawthorn fruit and hawthorn leaves can play a synergistic effect.3.The expression of AMPK?,p-AMPK?,SREBP-1 and ACC? in liver were determined by Western blotting to investigate the mechanism of different extracts of hawthorn in reducing blood glucose and lipid.Western blotting results show that compared with the model group,each administration medicine can increase the p-AMPK?/AMPK?,decrease SREBP-1,ACC? protein expression level,and the combination of hawthorn leaves flavonoids and hawthorn fruit organic acids is better than other medicine,suggesting that the hawthorn extracts can regulate glucose and lipid metabolism by activating AMPK?/SREBP-1/ACC? signaling pathway.Conclusion: hawthorn leaves flavonoids and hawthorn fruit organic acids have a certain effect on the prevention and treatment of glucose and lipid metabolism disorders.The mechanism may be related to AMPK?/SREBP-1/ACC? signal pathway,and the effect of fruit combined with leaves is better than alone.
