Glutaraldehyde-Polymerized Porcine Hemoglobin(pPolyHb)is an erythrocyte substitute with oxygen carrying/releasing oxygen function.The previous research was mainly used to treat acute anemia.In the experiment,Adenine-induced CKD anemia rat was used as an animal model.By measuring the anemia-related indexes and renal function parameters of rats after pPolyHb administration,it was investigated whether pPolyHb could correct chronic kidney disease(Chronic kidneydisease,CKD)anemia.Methods: Rats were given gavage of adenine at 250 mg/kg at different times.The optimal CKD anemia animal modelling scheme was selected according to the state and survival rate of the rats and the anemia judgment index.After modeling,rats were injected with three doses of 1,5,7 mg/kg pPolyHb and 5 mg/kg p Hb and 150 U/kg ESA for 4 weeks.Then the state,weight and survival rate of the rats were detected to investigate the effect of pPolyHb on the hematopoietic system of CKD anemia rats by detecting the Hb concentration,RBC content and Ret# of rats.By detecting the levels of IL-1?,IL-6 and TNF-? in rats and the content of hematopoietic promoting factor EPO by Elisa to explore the effect of pPolyHb on hematopoietic related factors in anemia rats.The level of Scr and BUN in rats was determined by biochemical analyzer analysis and the kidney index was calculated.Results:(1)After administration of pPolyHb(5 and 7 mg/kg)to anemia rats with CKD,the survival rate increased compared with the model group and the rats gained weight and were in better condition.At the end of the experiment,the Hb concentration,RBC content,and Ret# returned to normal levels and the anemia was corrected.(2)5 and 7 mg/kg pPolyHb decreased the levels of serum IL-1?,IL-6 and TNF-? in CKD anemia rats.The inhibitory effect of inflammatory factors on the hematopoietic system is slowed down and the content of EPO in rat serum is increased to normal levels.(3)5 and 7 mg/kg pPolyHb can reduce the abnormally elevated Scr and BUN concentration in CKD anemia rats to some extent.The accumulation of toxic substances in rats has been reduced.(4)After injection of positive drug ESA,the RBC content and Ret# of rats were significantly higher than other groups and the survival rate was lower than that of 5 and 7 mg/kg pPolyHb.Conclusion: The experiment shows that 5 and 7 mg/kg pPolyHb has a certain hematopoietic stimulation effect on adenine-induced chronic kidney disease anemia rats.PPoly Hb corrects anemia by increasing EPO levels in the body and increasing Hb concentration and RBC content in rats.PPoly Hb can not only reduce the inflammatory factors related to hematopoietic inhibition to a certain extent,but also reduce the levels of Scr and BUN in rats to slow down kidney damage.Moreover,pPolyHb increased the level of RBC in anemia rats more smoothly and steadily than the positive drug ESA,with higher survival rate and relatively better safety.
