Objective: Disease and disability are gradually becoming a threat to the health of all aspects of the world's population,and the aging of the world's population is increasing the number of people suffering from disease and disability sequelae,among which the number of patients with disc injury caused by various reasons is increasing day by day,resulting in a huge social and economic burden.In this experiment,we will explore the feasibility of acupuncture in the establishment of the rat tail intervertebral disc degeneration model,and explore the signal pathway of sIL-13R?2-Fc fusion protein in the inhibition of the process of intervertebral disc degeneration.Methods: the degeneration of the caudal vertebrae was induced by puncturing the caudal vertebrae of rats with acupuncture.The model of the degeneration of the caudal vertebrae of rats was established.The fusion protein of sIL-13R?2-Fc was injected into the model.The changes of collagen in the process of degeneration were observed by HE staining,Masson staining and Sirius red staining.The changes of GAG,HS,KS and HA in the process of degeneration were detected by ELISA to verify the changes of sIL-13R?2-Fc.The inhibition of fusion protein and the related signal pathway in this process were explored by molecular biology.Results: acupuncture can establish the model of the degeneration of the caudal intervertebral disc in rats;the application of sIL-13R?2-Fc in the model of the degeneration of the caudal intervertebral disc can improve the staining score of the intervertebral disc,and increase the content of proteoglycan,hyaluronic acid,chondroitin sulfate and keratin sulfate in the caudal intervertebral disc.It was found that PI3 K / Akt,peroxidase,TGF-? signaling pathway,Wnt signaling pathway and RAS / MEK / ERK signaling pathway were involved in the occurrence of disc degeneration.Conclusion: 1.The application of sIL-13R?2-Fc fusion protein in the model of intervertebral disc degeneration can improve the staining score of the intervertebral disc and increase the content of proteoglycan,hyaluronic acid,chondroitin sulfate and keratin sulfate in the caudal intervertebral disc tissue;2.The signal pathways involved in the inhibition of sIL-13R?2-Fc after injury are PI3 K / Akt,peroxidase,TGF-? signal pathway and wn signal pathway T signal pathway and RAS / MEK / ERK signal pathway;3.It was preliminarily confirmed that sil-13 r ? 2-fc had inhibitory effect on disc degeneration after injury.
