Atomic force nano-manipulation technology shows great advantages in measuring,characterizing and manipulating the samples on subcellular scales with the features of precision positioning and real-time monitoring of cell dynamics.The interaction between drugs and cells can be studied according to the cellular mechanical properties based on AFM(Atomic force microscope),which provides theoretical and experimental bases for the drug efficacy evaluation.This thesis studies the effects of 5-fluorouracil and paclitaxel on the morphology and mechanical properties of A549 lung cancer cells in liquid environments.The cellular physical properties have shown significant changes through analyzing a large number of cellular morphology images and mechanical properties.The developed nano manipulation system and AFM probe-single cell interaction have been utilized to evaluate the mechanical properties of live cells.The model of AFM probe and cell interaction is established,and the changes of cellular physical properties are analyzed quantitatively after the treatment of lung cancer cells with 5-fluorouracil.Furthermore,we extract the geometry and texture features of images to study the scan imaging and improve the image quality,which shows the changes of the surface morphology of lung cancer cells before and after the drug treatment.Based on the experiment above,the optimized hybrid mechanical model of probe-cancer cell is established by adding the velocity variable.The dose response and the time response of the paclitaxel treated lung cancer cells are quantitatively studied.Simultaneously,the changes of cell morphology,roughness,adhesion force and Young's modulus are obtained to analyze the effects of paclitaxel treatment on lung cancer cells.The model is verified through the force curve of the live cells in different concentrations of the drugs.The results show that the changes of cellular morphology and mechanical properties are associated with the drug,and provide a new way for the drug efficacy evaluation and drug discovery.
