Objective Acute lung injury(ALI)/ Acute respiratory distress syndrome(ARDS)is a common clinical critical disease with a high fatality rate,which seriously threatens the life of critically ill patients and affects their quality of life.To date,no drug has been shown to reduce mortality in patients with ALI / ARDS.ALI is essentially an inflammatory response to the lungs,so anti-inflammatory is the subject of ALI treatment.Gegen Decoction has been shown to have anti-inflammatory effects.The purpose of this experiment is to explore the effect of Ge Gen decoctionon acute lung injury,in order to provide new ideas for the study and application of traditional Chinese medicine in ALI/ARDS,and to provide a good theoretical basis for the application of Ge Gen decoctionin the treatment of clinical ALI patients in the future.Methods1.Twenty-four 8-week-old SD rats,all male,were controlled at 200± 20 g.2.Grouping: control group,models group,Gegen group,8 rats in each group.3.Decoction of Chinese medicine: Pueraria lobata 20 g,ephedra 15 g,twig 15 g,peony 15 g,ginger 15 g,jujube 10 g,licorice 10 g4.Gavage: The rats of Gegen group were gavaged according to the Chinese herbal medicine 900 mg / 100 g,and the models group was given the same dose of saline for 5 consecutive days.5.Modeling: On the fifth day,the models group and the Gegen group: 2.5 mg / kg tail vein injection of lipopolysaccharide(LPS),the control group: tail vein injection of the same volume of saline.6.All rats were sacrificed 48 h after modeling,and lung tissue and alveolar lavage fluid(BALF)specimens were collected.7.Pathological sections and pathological scores of rat lung tissue.8.Detection of wet / dry ratio of rat lung tissue.9.The total protein concentration of BALF in rats was detected by Coomassie brilliant blue method.10.Total cell count of rat BALF.11.Enzyme-linked immunosorbent assay(ELISA)was used to detect the expression of IL-6 in BALF in rats.12.Detect myeloperoxidase(MPO)activity in rat lung tissue.13.Western blot(WB)was used to detect the expression of Toll-like receptors 4(TLR4)in rat lung tissue.Result1.Compared with the control group,the pulmonary hemorrhage of the models group was severe,and congestion points appeared.Compared with the models group,the bleeding of lung tissue was reduced in Gegen Decoction group.2.Compared with the control group(0.01±0.01),the pathological score of the models group(0.26±0.02)increased(P <0.05).Compared with the models group,the pathological score of Gegen Decoction group(0.09±0.01)was reduced(P <0.05).3.Compared with the control group(4.51±0.05),the ratio of wet /dry weight of lung tissue in the models group(5.20±0.09)was increased(P <0.001).Compared with the models group,the ratio of wet / dry weight of lung tissue in Gegen Decoction group(4.86 ± 0.09)was reduced(P <0.001).4.Compared with the control group(0.401±0.11),the total protein concentration of BALF in the models group(1.11±0.11)increased(P<0.001).Compared with the models group,the total protein concentration of BALF in Gegen Decoction group(0.78±0.10)decreased(P <0.001).5.Compared with the control group(61.74 ± 24.58),the total number of BALF cells in the models group(229.56±10.50)increased(P <0.001).Compared with the models group,the total number of BALF cells in Gegen Decoction group(157.50±23.89)was reduced(P <0.001).6.Compared with the control group(19.73±3.33),the IL-6 level of BALF in the models group(73.85±3.26)and Gegen Decoction group increased(P <0.001).Compared with the models group,the IL-6 level of BALF in Gegen Decoction group(53.64±5.89)was reduced(P <0.05).7.Compared with the control group(0.22±0.09),the MPO activity level in the lung tissue of the models group(1.49±0.08)increased(P<0.001).Compared with the models group,the MPO activity level in the lung tissue of Gegen Decoction group(0.71 ± 0.11)was reduced(P<0.001).8.Compared with the control group(0.47±0.06),the expression of TLR4 protein in the models group(0.95 ± 0.08)was significantly increased(P <0.05).Compared with the models group,the expression of TLR4 protein in Gegen Decoction group(0.82 ± 0.09)decreased(P<0.05),indicating that Gegen Decoction can inhibit TLR4 protein expression.Conclusion Gegen Decoction can reduce LPS-induced acute lung injury and has anti-inflammatory effects,which may be related to reducing the expression of TLR4.
