Objective: To detect the levels of serum interferon-?(IFN-?),elastase(NE)and myeloperoxidase(MPO)in patients with SLE,to explore the correlation of IFN-?,NE and MPO levels with disease activity,serological indexes and clinical manifestations,and to understand the role of IFN-? and neutrophil extracellular traps(NETs)levels in the pathogenesis of SLE and the relationship between IFN-? and NETs,so as to provide more comprehensive detection indexes and treatment strategies for clinical diagnosis and treatment.Methods: 98 cases of SLE and 63 cases of healthy control group were collected.The peripheral venous blood was collected on admission and the serum was obtained after centrifugation.The levels of serum IFN-?,NE and MPO were measured by Enzyme-linked immunoassay(ELISA)method between SLE patients and healthy controls.Statistical methods were used to analyze the differences of IFN-?,NE and MPO levels between SLE patients and healthy controls,and to analyze the correlation of IFN-?,NE and MPO levels with disease activity,serological indexes and clinical manifestations in SLE patients.At the same time,the correlation between the level of IFN-? and the level of NETs in patients with SLE was analyzed.Results:1.The ELISA detection results of IFN-?,NE and MPO in the SLE patient group and the healthy control group: the levels of IFN-?,NE and MPO in the SLE patient group were significantly higher than those in the healthy control group(p<0.05).2.The relationship between the levels of IFN-?,NE,MPO and disease activity in patients with SLE: the levels of IFN-?,NE,MPO in patients with SLE were positively correlated with systemic lupus erythematosus disease activity index(SLEDAI)score(r=0.461,P=0.000;r=0.268,P=0.008;r=0.203,P=0.045).3.The relationship between the levels of IFN-?,NE,MPO and serological indexes in patients with SLE: In patients with SLE,the level of serum IFN-? was negatively correlated with serum complement C3 and Hemoglobin(Hb)(r=-0.204,P=0.044;r=-0.213,P=0.035).The level of serum IFN-? in patients with SLE was positively correlated with erythrocyte sedimentation rate(ESR),24 hour urinary protein and antids DNA antibody level(r=0.217,P=0.032;r=0.262,P=0.009;r=0.225,P=0.026),but not with C-reactive protein(CRP),platelet,anticardiolipin antibody,anti-RNP antibody,anti-sm antibody and anti-ribosomal p protein antibody(p>0.05).The level of serum NE in patients with SLE was not related to Hb,24 hour urinary protein,anti ds DNA antibody,serum complement C3,ESR,CRP,platelet,anticardiolipin antibody,anti-RNP antibody,anti-sm antibody and anti-r RNP antibody.The level of serum MPO in patients with SLE was positively correlated with anti-ds DNA antibody(r=0.208,P=0.04),but not with Hb,24 hour urinary protein,serum complement C3,ESR,CRP,platelet,anticardiolipin antibody,anti-RNP antibody,anti-sm antibody and anti-r RNP antibody(p>0.05).4.The relationship between the levels of IFN-?,NE,MPO and clinical manifestations in patients with SLE: the level of serum IFN-? in patients with SLE was positively correlated with clinical manifestations such as blood system,urinary system,arthritis,rash,photosensitivity and so on(r=0.205,P=0.043;r=0.237,P=0.019;r=0.252,P=0.012;r=0.215,P=0.034;r=0.235,P=0.020).It was not related to mental nervous system(p>0.05).The level of serum NE in patients with SLE was positively correlated with the clinical manifestations of rash(r = 0.279,P=0.050),but not with blood system,urinary system,arthritis,photosensitivity,mental nervous system and so on(p>0.05).The levels of serum MPO in patients with SLE were not related to blood system,urinary system,arthritis,photosensitivity,mental nervous system and skin rash(p>0.05).5.Correlation among serum IFN-?,NE and MPO levels in SLE patients: the results showed that NE and MPO in SLE patients were positively correlated(r=0.234,p=0.020),and IFN-? was positively correlated with NE and MPO respectively(r=0.245,p=0.015;r=0.273,p=0.007).Conclusion:1.The serum levels of IFN-?,NE and MPO in patients with SLE were higher than those in normal group,suggesting that the levels of IFN-? and NETs matter in the pathogenesis of SLE.2.The levels of IFN-?,NE and MPO in patients with SLE were positively correlated with SLEDAI score,suggesting that the levels of IFN-?,NETs were related to the disease activity of SLE.3.The relationship between the levels of serum IFN-?,NE and MPO and the measured serological indexes in patients with SLE suggests that both IFN-? and NETs may be involved in the pathological process of SLE.4.The relationship between the levels of serum IFN-?,NE and MPO and the clinical manifestations of each system in patients with SLE suggests that the levels of serum IFN-?,NE and MPO may play an important role in the pathogenesis of SLE,and may be related to the pathological process of clinical manifestations of each system.5.There was a positive correlation between NE and MPO in patients with SLE,suggesting that both NE and MPO could reflect the level of NETs,and they were consistent.IFN-? was positively correlated with NE and MPO respectively,indicating that IFN-? and NETs may be related in the pathogenesis of SLE.
