Objective: This study explored the possible effects of siRNA HO-1 on the function of inorganic arsenic in suppressing dendritic cells,and further explored the role of HO-1 in inorganic arsenic-mediated immune suppression,so as to provide theoretical basis for the prevention and treatment of arsenic-related diseases.Methods: 1.Induction culture of mouse BMDCs.2.Detection indicators and methods:(1)HO-1 silencing BMDCs were established by gene silencing.(2)Western blot was used to detect the HO-1 protein content in DC.(3)The content of related cytokines in DC was detected by real-time quantitative PCR.Results: 1.HO-1 silencing reduced the expression of HO-1 protein and mRNA in BMDCs exposed to inorganic arsenic.After BMDCs were transfected with Control siRNA and HO-1 siRNA,compared with LPS group in si Ctr group and si HO-1 group,the expression of HO-1 protein and mRNA in As + LPS group in si Ctr group and si HO-1 group The level increased;compared with the As + LPS group in the si Ctr group,the HO-1 protein and mRNA expression levels in the si HO-1 group decreased significantly.The differences were statistically significant(P < 0.05).2.HO-1 silencing increased mRNA expression of co-stimulatory molecules and chemotactic adhesion factors in BMDCs exposed to arsenic.Compared with the As + LPS group in the si Ctr group,the mRNA transcription levels of Cd86,Cd40,Ccr5 and Ccr7 in the As + LPS group in the si HO-1 group increased,and the difference was statistically significant(P < 0.05).3.HO-1 silencing increased mRNA expression of pro-inflammatory factors in arsenic-exposed BMDCs.Compared with the As + LPS group in the si Ctr group,the expression of the mRNA transcription level of Il-1? in the As + LPS group in the si HO-1 group increased.The difference was statistically significant(P < 0.05).4.HO-1 silencing increased the expression of Th1,Th2,Th17 and Treg cytokine mRNA in BMDCs exposed to arsenic.Compared with the As + LPS group in the si Ctr group,the mRNA expression levels of pro-inflammatory factors,Th1,Th2,Th17,and Treg cytokines in the As + LPS group in the si HO-1 group were all increased.The differences in the factors Il-12 b,Il-4,Il-6,Il-23 a and Il-10 were statistically significant(P < 0.05).Conclusion: 1.HO-1 silencing can increase the mRNA expression of arsenic-exposed BMDCs co-stimulatory molecules CD86,CD80 and chemotactic adhesion factors Ccr5,Ccr7.2.HO-1 silencing can increase mRNA expression of pro-inflammatory factor Il-1? in BMDCs exposed to arsenic.3.HO-1 silencing can increase the m RNA of Th1 cytokines Il-12 b,Th2 cytokines Il-4,Th17 cytokines Il-6 and Il-23 a,Treg cytokines Il-10 in exposed BMDCs expression.
